Study on D-galactose-induced Animal Model of Kidney-deficiency Polyuria

摘要 【目的】探索D-半乳糖(D-Ga1)致肾虚多尿动物模型的合适剂量,为该种模型应用于衰老所致肾虚多尿证的研究提供参考。【方法】将筛选后的SD大鼠随机分为4组,分别为正常对照组,D-半乳糖125、150、175 mg.kg-1.d-1剂量组;采用D-半乳糖连续注射8周,停止注射后继续观察1周,以24 h总尿量及水负荷5 h后尿量为直接观测指标,同时采用尿动力仪检测膀胱储尿及排尿功能以探索D-Ga1致肾虚多尿动物模型的合适剂量。【结果】从各组大鼠24 h总尿量及水负荷5 h后尿量观察,D-Ga1 150 mg.kg-1.d-1剂量组增加尿量作用更显著。从尿动力参数观察,D-Ga1 150 mg.kg-1.d-1剂量组对大鼠膀胱漏尿点压(BLPP)、腹压漏尿点压(ALPP)、顺应性(BC)、最大膀胱排尿压(MVP)、残余尿量(PVR)、排尿效率(EV)的影响更显著,且其改变趋势与衰老动物尿动力学的变化相符。【结论】D-Ga1 150 mg.kg-1.d-1剂量组在对尿液的生成、膀胱储存及排泄功能的影响方面更符合衰老所致肾虚多尿动物模型的制备。 Objective To optimize the appropriate dosage of D-galactose (D-Gal) for establishing kidney- deficiency polyuria animal model, and to offer references for the application of this animal model in studying aging induced kidney-deficiency polyuria. Methods The rats were randomly divided into normal control group and three D-Gal groups (in the dosages of 125, 150 and 175 mg.kg-1.d-1 respectively) . The rats were injected with D-Gal for 8 continuous weeks. During one week of D-Gal suspension, 24-hour urine volume, and the urine volume after water load for 5 hours were monitored. At the same time, the bladder functions of reserving urine and micturition were detected with urine dynamometer. Results Of the three D-Gal dosages, 150 mg.kg-1.d-1 D-Gal showed obvious effect on increasing 24-hour urine volume and the urine volume after water load for 5 hours, also showed remarkable effect on urine kinetic indexes including bladder leak point pressure (BLPP), abdominal leak point pressure (ALPP), bladder compliance (BC), maximum voidingpressure (MVP), post-voidresidual urine volume (PVR), and efficiency of voiding (EV) . The action trend accorded with the changes of urine kinetics in aging animal. Conclusion It is shown that 150 mg.kg-1.d-1 D-Gal is the appropriate dosage to make the aging-induced kidney-deficiency polyuria animal model.
