摘要
目的根据九味头痛片临床疗效及其功能主治,对其进行药效学及作用机制的实验研究。方法镇痛实验采用小鼠扭体、热板两种实验方法,同时皮下注射硝酸甘油建立大鼠偏头痛模型,观察九味头痛片对大鼠脑血流量、血液流变学指标的影响。结果九味头痛片明显减少小鼠因醋酸刺激所致扭体的次数,并呈现一定的量效关系;能显著提高小鼠热刺激所致疼痛的痛阈值,且作用持续时间较长(在4h时作用最强)。经九味头痛片治疗后大鼠脑血流量得到明显改善,全血低切、高切还原粘度显著下降。结论九味头痛片能显著提高小鼠的痛阈值,对醋酸所致小鼠的疼痛有显著缓解作用,该药物对偏头痛的镇痛作用可能因其能够降低血液粘度,改善脑血流,促进脑血液循环所致。
Objective To study the pharmacodynamics and mechanism of Jiuwei headache tablet (JWTTP) at relieving pain in mice. Methods Two experimental methods (mice writhing and hot plating) were utilized to evaluate the analgesic effect of JWTTP. Mouse models of migraine were established by subcutaneous injection of nitroglycerin. The influence of JWTTP on mouse cerebral blood flow and hemorheology was examined. Results JWTTP significantly reduced the number of writhing in- duced by acetic acid stimulation and the effect was in a dose-dependant manner. JWTTP significantly elevated the pain threshold of the mice experiencing hot stimulation and markedly increased the percentage of the pain threshold. Moreover,the analgesic effect of JWTTP lasted a long time and was strongest at 4 h after treatment. The cerebral blood flow was significantly improved in mice after JWTTP treatment and the blood viscosity was obviously decreased. Especially,the whole blood low-shear reduced viscosity and erythrocyte aggregation index were reduced remarkably. Conclusion JWTTP significantly elevated the pain threshold of the mice and relieved the pain induced by acetic acid stimulation in mice. The mechanism of JWTTP at alleviating the migraine pain may be related to lowering blood viscosity, improving cerebral blood flow and eventually promoting cerebral blood circulation.
