摘要 目的副流感病毒兰州株的分离与鉴定。方法应用鸡胚和人胚肺成纤维突变细胞株分离病毒 ;应用血凝和血抑以及近年来多株流感国际和国家株 (H1N1亚型 6株、H3N2亚型 2株、B型 4株 )作抗原抗体交互试验 ;还用 4类主要呼吸道病毒单克隆抗体间接免疫荧光法与 2株兰州株的病毒抗原基质片作特异性交叉反应。结果 2株兰州株在鸡胚中培养血凝效价滴度高达 1:5 12 (+ + )。它们与流感病毒甲型 (H1N1和 H3N2 )以及乙型流感抗血清均无血抑反应。它们的血凝素抗原及血清与 12株甲和乙型流感病毒株无免疫血抑交互反应。但 2株流行株抗原片可和副流感病毒 2型单克隆抗体发生较强反应 ,在人胚肺细胞中培养有明显细胞融合病变。结论 2株流行株虽具有高滴度血凝效价 ,但不是流感病毒株 ,根据单克隆抗体荧光染色和流行株在人胚肺细胞中的病变 ,初步确认为副流感病毒 ObjectiveTo identify two epidemic strains of parainfluenza virus isolated from secreta of two soldiers in Lanzhou. Methods Viruses were isolated by using chicken embryo and mutant lung fibroblast (Z HL 16C) of human embryo. The viruses isolated were assayed by hemagglutination (HA) test,hemagglutination inhibition (HAI) test and antigen antibody cross test with several international and national influenza strains (4 substrains of H1N1,2 substrains of H3N2,4 strains of Flu B).Virus antigen slides of them were also assayed by specific cross test with monoclonal antibodies(McAb) against 4 main respiratory track viruses.ResultsHA titer of two Lanzhou strains cultured in chicken embryo was high up to 1∶512(++). The results of HAI test were negative when assayed with anti sera of Flu A (H1N2 and H3N2) and FluB viruses.Negative results were also obtained in antigen antibody cross test with 12 strains of FluA and FluB viruses.However,strong fluorescence were obtained when antigen slides of two Lanzhou strains were tested with PIV 2 McAb. Pathological changes of cell fusion were obvious in Z HL 16C in which two Lanzhou strains were cultured. Conclusion The viruses isolated were not influenza viruses in spite of their high HA titer. We identified preliminarily them as PIV 2 type by results of McAb fluorescent staining and pathological changes in Z HL 16C.
