摘要
目的探讨慢性胃炎(CG)病人胃黏膜组织与血清中细胞因子白细胞介素(IL)-17、IL-6和转化生长因子β1(TGF-β1)的表达及其对CG发生发展的可能作用。方法选取我院CG病人141例,按有无幽门螺杆菌(Hp)感染分为Hp(+)组和Hp(-)组。对照组50例,Hp均为阴性,选取其中26例作为组织学对照。采用组织切片染色方法和胃黏膜组织快速尿素酶试验来判定有无Hp感染;采用ELISA法检测受检者血清中IL-17、IL-6、TGF-β1的浓度;采用免疫组化法测定胃黏膜组织中IL-17、IL-6、TGF-β1的表达。结果 CG病人血清和胃黏膜组织中IL-17、IL-6、TGF-β1的表达均较正常对照组增强(F=10.47~25.64,q=3.14~10.04;χ2=20.03~35.35;P<0.05),IL-17、TGF-β1在血清和胃黏膜组织中的表达均与炎症程度呈正相关,IL-6在胃黏膜组织中的表达与炎症程度呈正相关。IL-17、IL-6、TGF-β1在Hp(+)组病人血清和胃黏膜组织中的表达均较Hp(-)组增强(t=4.45~7.00,χ2=14.29~22.41,P<0.05、0.01)。结论在CG的发生发展过程中细胞因子IL-17、IL-6、TGF-β1可能参与其中发挥作用。
Objective To explore expressions of cytokines IL-17,IL-6,and TGF-β1 in chronic gastritis(CG) and potential effects of the expressions on occurrence and development of the disease.Methods According to whether or not with Helicobacter pylori(Hp) infection,141 CG patients were divided into Hp-positive group and Hp-negative group.Fifty CG patients with Hp negative were served as control group,of whom,26 as histology-control.Using a tissue section staining method and gastric mucosal tissue rapid urease test to determine whether or not the mucosa was infected with Hp;ELISA to detect serum concentration of IL-17,IL-6,and TGF-β1 in the subjects;and the expressions of IL-17,IL-6 and TGF-β1 in gastric mucosa were measured by immunohistochemical technique.Results The expressions of IL-17,IL-6,and TGF-β1 in serum and gastric mucosa of the patients were significantly higher than that in the normal control(F=10.47-25.64,q=3.14-10.04;χ2=20.03-35.35;P<0.05),that of IL-17 and TGF-β1 in serum and gastric mucosa were positively correlated with the degree of inflammation,and that of IL-6 was positively correlated with the degree of inflammation in gastric mucosa.The expressions of the three cytokines in Hp-positive group in both serum and gastric mucosa were higher than that in Hp-negative group(t=4.45-7.00;χ2=14.29-22.41;P<0.05,0.01).Conclusion Cytokines IL-17,IL-6,and TGF-β1 may be involved in the occurrence and development of chronic gastritis.
