摘要
目的研究大鼠失神经支配眼轮匝肌超微结构及胶原纤维含量变化,为选择最佳修复时机和制定治疗方案提供理论依据。方法制作大鼠失神经支配眼轮匝肌模型,分正常组,术后1、3d,1、2、4、6周组标本,透射电镜观察超微结构,Masson染色检测胶原纤维与肌细胞面积比的变化。结果 (1)不同时间组肌组织中胶原纤维与骨骼肌细胞面积比总体有差异(P<0.05),并且随着失面神经时间的延长呈递增趋势;(2)失神经肌组织超微结构由正常的结构功能,随时间的延长,肌纤维膜系统肿胀、排列紊乱、核固缩、线粒体肿胀变性、溶酶体增生等并进一步加重。结论随着失神经时间的延长,胶原纤维增多、肌肉萎缩越明显,肌组织失神经损伤时间越长,损伤越严重,应在发现损伤后尽早修复,以恢复面神经支配功能。
Objective It was aimed at the rats orbicularis oculi muscle cell uhrastructure and collagenous fiber changes of facial-nerve denervation in order to provide a theoretical basis for the best timing of facial-nerve surgical repair and making the treatment plan. Methods Rat models of orbicularis oculi muscle cell of facial-nerve denervation at different periods (ld,3d, lw,2w,4w,6w) were made and the orbicularis oculi muscle cell and collagenous fiber were observed by transmission electron microscope observation and Masson dyeing analysis at different periods. Results (1) In the different periods the area of collagen fibers and skeletal muscle cells have difference with the overall group (P 〈 0.05) and the area showed a trend of increased during the duration of facial nerve innervation loss;(2) With the extension of time,muscle tissue ultrastructure of innervation loss changed from normal structure system to Muscle fiber membrane system was swelled,disordered arrangement, nuclear pyknosis,mitochondria swelling,lysosome hyperplasia and be worse. Conclusion With the growing of time of innervation loss,collagen fibers increased,muscle atrophied obviously.The longer the loss of muscle tissue innervation injury,the more serious the damage.So we should repair as soon as possible after detection of the injury,to resume the function of facial nerve dominated.
