摘要
使用热灭活的卡介苗刺激培养的人肺腺上皮细胞(SPC-A-1),利用逆转录聚合酶链反应(RT-PCR)法和Northern杂交分析,证实了卡介苗能诱导SPC-A-1细胞β-防御素-1基因(hBD-1)的增强表达。卡介苗菌量和刺激时间与SPC-A-1细胞hBD-1基因表达量的关系曲线显示,hBD-1 mRNA 在人肺腺上皮细胞的增强表达在一定范围内表现为具刺激剂量和时间依赖关系。实验为开发增强粘膜抗菌肽基因表达的免疫增强剂以防治粘膜感染的研究打下了基础。
Using reverse-transcription PCR (RT-PCR) and Norther n blot analysis, the present authors found an enhanced expression of the human β-defensin 1 (hBD-1) gene in the SPC-A-1 cells chanllenged with heat-kille d M. Bovis Bacille Calmette-Guerin (BCG). The enhanced expression of hBD1 mRNA was dose- and time-dependent. This result indicated a significant progress in the work of developing a new antimicrobial strategy that enhances mucosa ant ibiotic peptide expression for the prevention and treatment of mucosal infectio ns.
