苦参碱抑制血小板衍生生长因子诱导的小鼠皮肤成纤维细胞增殖 被引量:40

摘要 目的 :探讨苦参碱对血小板衍生生长因子 - BB(PDGF- BB)诱导的小鼠成纤维细胞增殖的影响。方法 :取新生 ICR小鼠皮肤细胞培养成纤维细胞。用四甲基偶氮唑盐 (MTT)法和细胞计数法分析细胞增殖情况。结果 :5 0~ 2 5 0 μg/ ml的苦参碱能剂量依赖地降低血清刺激的细胞增殖 ,撤除苦参碱后抑制作用能完全逆转。苦参碱 (10 0~ 5 0 0 μg/ ml)亦能剂量依赖地抑制PDGF- BB诱导的细胞增殖。结论 :苦参碱显示了很强的抗小鼠皮肤成纤维细胞增殖的作用 ,可能通过抑制 PDGF- BB诱导增殖作用来介导的。提示苦参碱可能对皮肤纤维化疾病有潜在的预防和治疗作用。 Objective: To test the effect of matrine on platelet derived growth factor BB (PDGF BB) induced proliferation of mouse skin fibroblasts. Methods: Mouse skin fibroblasts were obtained from newborn ICR mice and propagated in vitro . Proliferation of cells was analyzed by mitochondrial reduction of tetrazolium salt MTT to formazan and actual cell count. Results: Matrine (50 250 μg/ml) caused reduction of serum driven cell growth dose dependently. Growth inhibition was totally reversed after removal of the drug. Matrine also produced inhibition of PDGF BB induced cell growth dose dependently. Conclusion: Matrine exhibits potent anti proliferation effect against mouse skin fibroblast. This effect appears to be mediated by a reduction of PDGF BB induced growth. These results suggest that matrine might have therapeutic implication in skin fibrosis. [
