The relationship between ANGPTL4 gene expression, angiogenesis and progression of gastric cancer

摘要 目的探讨ANGPTL4 mRNA的表达和血管生成与胃癌发展的关系。方法选择50例胃癌患者组织标本作为实验组,10例正常胃黏膜标本作为对照组,采用原位杂交技术检测ANGPTL4 mRNA表达,免疫组化染色法检验微血管密度,并分析其与胃癌临床病理学特征的关系。结果 ANGPTL4 mRNA阳性率及微血管密度MVD计数均为胃癌组织>癌旁组织>正常胃黏膜,差异有统计学意义(P<0.01)。经Pearson相关分析,ANGPTL4mRNA阳性率与MVD计数呈正相关(P<0.05)。胃癌组织中ANGPTL4阳性表达率与MVD计数与年龄、性别无关,与组织病理分级、TNM分期、浸润深度及淋巴结转移密切相关(P<0.05),分别与组织病理分级、TNM分期、浸润深度呈正相关(P<0.05)。结论胃癌组织中ANGPTL4mRNA高表达可能促进新生血管形成,肿瘤细胞浸润和转移。 Objective To explore the relationship between ANGPTL4 mRNA expression, angiogenesis and progression of gastric cancer. Methods The tissue specimens from 50 gastric cancer patients were selected as experiment group, while the normal gastric mucasa from 10 healthy people was chosen as control group. The ANGPTL4 mRNA expres sion was tested by in-situ hybridization. The mierovessel ing. The relationship between these parameters and clini density (MVD) was examined by Immunohistochernistry stain- copathologie features was analyzed. Results The positive rate of ANGPTL4 mRNA and MVD count all displayed in gastric cancer tissue 〉 para-carcinoma tissue 〉 normal gastric mucosa with a statistical difference (P 〈 0.01), By Pearson correlation analysis, the research found a positive correlation between the positive rate of ANGPTL4 mRNA and MVD count (P 〈 0.05). In gastric cancer tissues, the positive rate of ANGPTL4 mRNA and MVD count were not relatsd with age or gender, but in close relationship with histopathological grading, tumor-node-metastasis (TNM) staging, infiltration depth, and lymphatic metastasis (P 〈 0.05). Conclusion The high expression of ANGPTL4 mRNA in gastric tissue may boost up neovascularization, and infiltration and metastasis of tumor cells.
