rhEGF凝胶对兔皮肤Ⅲ度烧伤创面TNF-α水平的影响 被引量:3

摘要 目的观察重组人表皮生长因子(rhEGF)凝胶治疗兔皮肤Ⅲ度烧伤的作用及创面组织肿瘤坏死因子-α(TNF-α)水平的变化。方法铜棒烫伤致兔腹背部皮肤Ⅲ度烧伤,切痂后治疗侧用rhEGF凝胶涂抹创面,自身对照侧涂等体积生理盐水,肉眼观察两侧创面修复情况,并测量未愈合创面面积,HE染色后进行病理组织学检查,ELISA法检测组织TNF-α水平。结果治疗第3、18天,rhEGF治疗侧创面愈合率显著高于对照侧(P<0.05);治疗第3天,rhEGF治疗侧创面组织红肿明显较对照侧轻。病理学检查显示,rhEGF侧组织红肿明显较对照侧轻,炎细胞浸润也较少,且创面组织中TNF-α含量明显低于对照侧(P<0.01)。结论 rhEGF凝胶可明显促进兔皮肤Ⅲ度烧伤修复,与其在炎症期降低创面组织TNF-α水平,减轻炎症反应有关。 Objective To observe the effects of the recombinant human epidermal growth factor (rhEGF) gel on skin third degree burn in rabbits and the level of tumor necrosis factor-α (TNF-ct)in the wound tissue. Methods The skin third degree burn of front and rear part in the New Zealand rabbits was made by ambustion of copper bar. The rhEGF gel was smeared on the wound of the treatment side after surgical removal of the scar and equal - volume of physiological saline was smeared on the control side. The process of wound restoration was observed and unhealthy wound area was measured. HE Stai- ning of wound tissue was performed for histological examination. TNF-α level was detected using enzyme - linked immunosorbent assay. Results The wound healing rate of rhEGF treatment side was notably higher than that in the control side in the third and 18th days after treatment (P 〈 0.05 ). In the third day after treatment,red swelling in the rhEGF treatment side was markedly slighter than that is the that in the control side. histological examination showed that the milder edema, less inflammatory ceils and lower content of TNF-α in rhEGF treatment side were detected than those in the control side. Conclusion rhEGF gel could promote the repair of skin third degree burn in rabbits and this effect might be associated with the decreased TNF-α level of wound tissue and the reduced inflammatory reaction in inflammation phase.
