摘要
目的 :观察长期高脂饮食对小鼠学习记忆功能及情绪的影响。方法:将14只C57BL/6J成年小鼠随机分为2组:正常对照组(NC组)和长期高脂饮食组(HF组)。NC组无任何处理,HF组以40%高脂饲料饲养8周,之后行Morris水迷宫、八臂放射迷宫检测高脂饮食对小鼠学习记忆功能的影响,行开场实验、高架十字迷宫、强迫游泳和悬尾实验观察小鼠情绪及活动性的变化,同时,在造模期间观察体质量、空腹血糖、平均饮水量和饮食量等指标。结果:与NC组相比,HF组平均饮水量和饮食量增加,体质量明显增加;空腹血糖增高;Morris水迷宫提示HF组空间记忆受损,工作记忆未受损;八臂迷宫提示HF组参考记忆错误和工作记忆错误增多。开场实验提示HF组小鼠探索行为活跃,有焦虑行为;强迫游泳实验提示HF组有抑郁情绪和绝望行为。结论:长期高脂饮食可引起小鼠体质量和空腹血糖增高,可损伤小鼠的工作记忆和空间记忆,并引起小鼠活动性增加和焦虑抑郁行为。
Objective: To observe the effect of chronic high-fat diet on mice' learning, memory and mood Methods: Fourteen male adult C57BL/6J mice were randomly divided into 2 groups (seven in each): normal control group (NC group) and chronic high-fat diet group (HF group). NC group had no treatment at all, mice from HF group were fed with high-fat diet which has 40% calorie. Eight weeks later, spatial memory and working memory of these mice was tested by Morris water maze and 8-arm radial maze task, mood and activity was tested by open field test, elevated plus-maze test, forced swimming test and tail suspension test. Meanwhile, their weights in the first, fourth and the eighth week was recorded, fasting blood-glucose, the mean water uptake and food uptake were recorded too. Results: The water uptake, food uptake, weights and fasting blood-glucose of HF group increased greatly. In the Morris water maze task, mice in the HF group had longer escape latent period, and in the 8-arm (4-arm baited) radial maze, they had more reference memory error and more working memory error. In the open field test, the results indicated that mice in the HF group had more exploratory and anxiety behaviors. In the forced swimming test, the results indicated that the mice in the HF group were depressed. Conclusion: Longtime high-fat diet can increase mice' bodyweight and fasting blood-glucose, lead to anxiety-depression mood, meanwhile, their special reference memory and the working memory are impaired.
