摘要
目的研究壳聚糖-左氧氟沙星长效缓释敷料的体外细胞毒性、抗菌效果以及肿瘤坏死因子(TNF)-α作为评价抗菌效果指标的可行性。方法参照ISO10993-5:2009评价该敷料的细胞毒性;制作染菌创伤白兔模型15只并分为3组,空白对照组用凡士林油纱包扎,阳性对照组用纳米银创伤贴包扎,实验样品组用壳聚糖-左氧氟沙星长效缓释敷料包扎,于6h、1d、4d、7d,采用活组织培养法检测3组的菌落数,用双抗体夹心法检测TNF-α含量,与菌落数实验结果进行比较。结果该敷料体外细胞毒性反应为0级;实验样品组4个时点的菌落数均低于空白对照组,在6h、1d、4d时,实验样品组均低于阳性对照组(P<0.05);实验样品组的TNF-α含量在6h、1d、4d、7d时低于空白对照组,在6h、1d时显著低于阳性对照组(P<0.05)。结论壳聚糖-左氧氟沙星长效缓释敷料无细胞毒性,具有长效抗菌能力,TNF-α不能作为评价抗菌效果的指标。
Objective To study the in vitro cytotoxic dressing effect and antibacterial activity of microspheres containing levofloxacin, and the feasibility to use tumor necrosis factor (TNF)-a as the evaluate index. Methods The cell toxicity of the dressing was evaluated using ISO 10993-5:2009. The model with infected wounds was established in 15 white rabbits, The model rabbits were divided into 3 groups, blank control group (vaseline gauze dressing), the positive control group (nanometer silver paste trauma dressing) and experimental group (microspheres containing levofloxacin dressing). The bacterial colonies were detected using tissue culture assay in 6-hour, 1-day, 4-day and 7-day. The TNF-a content was detected by double antibody sandwich method, which was compared with the number of colonies. Results The cytotoxic reaction of the dressing was 0 colony in vitro. The number of colonies at 4 points was significantly lower in experimental group than that of blank control group (P 〈 0.05). The number of colonies was significantly lower at 6-hour, 1-day and 4-day in experimental group than that of positive control group (P 〈 0.05). The content of TNF-oL was significantly lower at 6-hour, l-day, 4-day and 7-day in experimental group than that in blank control group, and which was significantly lower at 6-hour, 1-day than that in positive control group (P 〈 0.05). Conclusion The wound dressing of microspheres containing levofloxacin has no cytotoxicity, and which has long-acting antibacterial ability. TNF-a can not be used as the evaluation index.
