TDZD-8对乙醛激活肝星状细胞中Wnt通路的作用研究

摘要 目的检测乙醛激活肝星状细胞中GSK-3β抑制剂TDZD-8对Wnt信号通路的作用。方法体外培养大鼠肝星状细胞株(HSC-T6),经乙醛刺激细胞活化后,分空白对照组和阻断组,阻断组加入TDZD-8(终浓度20μm/L)作用12h,MTT法检测细胞增殖;提取蛋白Western blotting检测Wnt途径下游产物β-catenin的表达状况及磷酸化水平变化。结果 MTT检测结果发现TDZD-8阻断GSK-3β之后乙醛刺激的肝星状细胞增殖高于对照组(P<0.05);Western blotting检测结果显示使用TDZD-8激活Wnt途径后,Wnt途径下游产物β-catenin表达无明显变化,但p-β-catenin水平下降。结论在乙醛激活的肝星状细胞中,TDZD-8能有效激活Wnt信号通路从而刺激肝星状细胞转分化为肌成纤维细胞。
