芪蛭益肺颗粒对慢性阻塞性肺疾病模型大鼠小气道上皮MMP-9、TIMP-1表达的影响 被引量:9

Influences of Qizhi Yifei Granules on expressions of MMP-9 and TIMP-1 in small airway epithelium in rats with chronic obstructive pulmonary disease

摘要 目的探讨芪蛭益肺颗粒改善慢性阻塞性肺疾病(COPD)模型大鼠小气道重塑作用与其干预基质金属蛋白水解酶-9(MMP-9)、基质金属蛋白水解酶抑制剂-1(TIMP-1)表达间的关系。方法 72只大鼠随机分为正常组、模型组和中药治疗组,采用香烟烟雾暴露及气道内滴入脂多糖的联合造模方法建立COPD大鼠模型,于实验第21天起,模型组每天灌胃生理盐水1次,中药治疗组每天灌胃芪蛭益肺颗粒1次,正常组正常喂养。各组大鼠于实验第30、40、60天分批处死,留取标本用免疫组化法测定气道上皮MMP-9、TIMP-1蛋白表达。结果模型组第30天MMP-9表达低于正常组,第60天高于正常组,差异均具有统计学意义(P<0.05),中药治疗组各期表达与模型组差异无统计学意义;模型组TIMP-1各期表达与正常组无差异,中药治疗组在第30天下调TIMP-1的表达,差异具有统计学意义(P<0.05)。模型组MMP-9/TIMP-1比值在第30、40天较正常组降低,中药治疗组在第30、40天该比值均较模型组提高,差异均具有统计学意义(P<0.05)。结论芪蛭益肺颗粒在40 d前可以通过上调MMP-9/TIMP-1比值干预COPD模型大鼠小气道重塑。 Objective To investigate the effect of Qizhi Yifei Granules on remodeling of small airway and expressions of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 ( TIMP-1 ) in rats with chronic obstructive pulmonary disease (COPD). Methods All rats (n = 72) were randomly divided into normal group, model group and treatment group. The rat model of COPD was established by using the method combining smoking fuming and intratracheal LPS instillation. From the 21~t day, model group was intragastrically given saline once a day, treatment group, Qizhi Yifei Granules once a day and blank group, normal feeding. All rats were killed on the 30th day, 40th day and 60th day in batches. The expressions of MMP-9 and TIMP-1 were detected by using immunohistochemistry technique. Results In model group, the expression of MMP-9 was lower on the 30th day and higher on the 60th day compared with normal group ( P 〈 0.05 ). The expression of MMP-9 had no statistical difference in treatment group compared with model group at all time points. In model group, the expression of TIMP-1 had no difference compared with normal group at all time points, and decreased in treatment group on the 30th day (P 〈 0.05 ). The ratio of MMP-9 to TIMP-1 decreased in model group compared with normal group on the 30th day and 40th day, and increased in treatment group compared with model group (P 〈 0.05 ). Conclusion Qizhi Yifei Granules can intervene the remodeling of small airway in rats with COPD through up-regulating the ratio of MMP-9 to TIMP-1 before 40 days.
