摘要
CHUP1(chloroplast unusual positioning 1)参与了叶绿体移动信号转导过程,对植物避免光伤害及提高光合效率方面具有重要作用,并且与植物抗寒性有密切关系。本研究以福州宦溪野生蕉(Musa spp.AB group)叶片为材料,采用同源克隆的方法,分离出CHUP1基因cDNA和DNA序列,GenBank登录号分别为JX123753、JX880084,命名为Mu-CHUP1。Mu-CHUP1 cDNA全长3 232 bp,ORF 2 931 bp,编码976个氨基酸。福州宦溪野生蕉Mu-CHUP1 cDNA序列与小果野蕉(M.acuminata,AA Group)全基因组测序中的CHUP1 cDNA序列的相似性为84.71%;福州宦溪野生蕉Mu-CHUP1 ORF的DNA序列含8个内含子、9个外显子,而小果野蕉全基因组测序中的CHUP1基因组DNA序列则有11个内含子、12个外显子,两者相差较大。生物信息学预测分析表明,Mu-CHUP1磷酸化位点多达62个,并且含有3个保守结构域,可能与其行使多样性的功能有关。
CHUP1 (chloroplast unusual positioning 1) was involved in the process of chloroplast signal transduction, which played important roles in avoiding photodamage and improving photosynthetic efficiency to plants, and it was closely related to cold-resistance in plants. In this report, the leaves of the wild banana (Musa spp., AB Group) from Huanxi Town were used as the materials for cloning CHUP1 genes by the homologous cloning method, and the cDNA and genomic sequences of CHUP1 were firstly isolated and named as Mu-CHUP1 (the accession numbers were JX123753 and JX880084 in GenBank, respectively) in Musa plants. The sequences of the full-length cDNA and ORF of CHUP1 were 3 232 bp and 2 931 bp, encoding 976 amino acids. The cDNA sequence similarity was 84.71% between Mu-CHUP1 and CHUP1 of M. acuminata (AA Group) from the genome database; Mu-CHUP1 had 9 exons and 8 introns, and CHUP1 of M. acuminata (AA Group) from the genome database had 12 exons and 11 introns, which had quite a few differences between them. Bioinformatic analysis showed that the phosphorylation sites of Mu-CHUP1 were as many as 62, and there were 3 conservative domains, which might be related to the functional diversity of Mu-CHUP1.
