摘要
将脱分化与再分化过程分开,探讨不同培养基和激素配比对黑暗条件下84K杨叶片及茎段外植体产生愈伤组织及愈伤组织诱导出芽的影响。结果表明,84K杨叶片产生愈伤组织的高效培养基为MS+KT 0.5 mg/L+2,4-D 0.2 mg/L;茎段为WPM+KT0.5 mg/L+2,4-D 0.1 mg/L。愈伤组织诱导出芽的高效培养基为WPM+TDZ 0.05 mg/L+6-BA 0.5 mg/L。为下一步获得高效的84K杨转基因体系提供一个高效受体系统。
Separating regeneration system into dedifferentiation and redifferentiation processes, we investigated the effects of different medium and hormones on callus induction under dark condition and adventitious buds from induced callus by using the leaves and stem segments of Poplar 84K as explants. The results showed that the efficient medium for callus induction from Poplar 84K leaves was MS+KT 0.5 mg/L+2,4-D 0.2 rag/L; for stem segments it was WPM+KT 0.5 mg/L+2,4-D 0.1 mg/L. The efficient medium for callus-induced adventitious buds was WPM+TDZ 0.05 mg/L+6-BA 0.5 mg/L. This study provided an efficient regeneration system, which would be beneficial for the establishment of an efficient transgenic system of Poplar 84K in the future.
