摘要
目的探讨反相高效液相色谱测定人体甲氧苄胺嘧啶血药浓度的方法与效果。方法选择Agilent1100反相高效液相色谱仪测定人体甲氧苄胺嘧啶的血药浓度。结果甲氧苄胺嘧啶的线性方程为Y=6.25×10-4X+1.08×10-2,线性范围为0.05~10.00μg/ml。甲氧苄胺嘧啶日内和日间精密度的相对标准偏差分别为4.4%和4.5%,提取回收率为92.1%,平均血药浓度为0.260μg/ml。结论采用反相高效液相色谱测定人体甲氧苄胺嘧啶血药浓度,方法简单、可行、灵敏度高,具有良好的回收率、准确度和精密度。
Objective To investigate the method and effect of reversed phase high-performance liquid chromatography (RP-HPLC) in the determination of human trimethoprim plasma concentration. Methods Agilent 1100 RP-HPLC instrument was selected to measure the concentration of human plasma trimethoprim. Results Trimethoprim linear equation Y = 6. 25 × 10 -4X + 1. 08 × 10-2 ,the linear range was 0. 05 -10. 00 μg/ml. The relative standard deviation of trimethoprim intraday and interday precision (RSD) were 4. 4% and 4. 5% respectively,the recovery rate was 92. 1% ,the average concentration in the plasma was 0. 260μg/ml. Conclusion The method that determining human trimethoprim plasma concentration with RP-HPLC is simple,feasible,high sensitivity,and has good recovery,accuracy and precision.
