南方红豆杉水提物协同厄洛替尼对人肺癌A549细胞COX-2、MMP-2表达的影响 被引量:7

摘要 目的研究南方红豆杉水提物协同厄洛替尼抑制人肺癌A549细胞及对其COX-2、MMP-2表达的影响。方法采用MTT法检测厄洛替尼对A549细胞增殖的抑制作用;采用MTT法检测不同浓度厄洛替尼联合南方红豆杉水提物干预后,人肺癌A549细胞的增殖情况;Western blot法检测不同浓度厄洛替尼联合南方红豆杉水提物干预后人肺癌A549细胞环氧酶-2(Cyclo Oxygenase-2,COX-2)、基质金属蛋白酶-2(Matrix Metallo Protein-ase-2,MMP-2)的蛋白表达量。结果厄洛替尼对A549细胞的增殖有抑制作用;低剂量厄罗替尼联合南方红豆杉水提物组抑制率明显高于标准剂量厄洛替尼组(P<0.05);Western blotting法显示两药联合组COX-2、MMP-2的蛋白表达明显低于相应单药厄洛替尼组(P<0.05),低剂量厄洛替尼联合标准剂量南方红豆杉水提物组COX-2、MMP-2蛋白表达明显低于标准剂量厄洛替尼组(P<0.05)。结论南方红豆杉水提物能协同厄洛替尼增强抑制人肺癌A549细胞增殖,疗效呈相加效果,下调COX-2、MMP-2蛋白表达为其可能机制。 Objective To study synergistic effect of aqueous extract Taxus chinensis combining Erlotinib on human lung carcinoma A549 cells and possible mechanisms. Methods The inhibitory effect of Erlotinib on human lung carcinoma A549 cells was tested by MTT method; The synergistic effect of Erlotinib combining aqueous extract Taxus chinensis on human lung carcinoma A549 cells was tested by MTT method; Western blot was used to detect the expression of COX-2, MMP-2 in human lung carcinoma A549 cells. Results Erlotinib has inhibitory effect on human lung carcinoma A549 cells,Inhibition ratio in group of lower Erlotinib combining Taxus chinensis are lower than that in standard Erlotinib group only (P 〈 0.05); Protein levels of COX-2, MMP-2 pointed out that Taxus chinensis combining Erlotinib had Syner gistic effect on human lung carcinoma A549 cells, and expression of COX-2, MMP-2 in group of lower Erlotinib combining Taxus chinensis are lower than that in standard Erlotinib group only (P 〈0.05). Conclu- sion The experimental results show that anticancer effects of Tasus chinensis vat. mairei combining Erlotinib on A549 cells may be associated with downexpression of COX-2, MMP 2. Aqueous extract Taxuschinensis may partly replaced the effect of Erlotinib.
