摘要
目的探讨七叶皂苷钠对急性脑出血(intracerebral hemorrage,ICH)后核转录因子(NF-κB)和水通道蛋白(AQP)动态表达的影响。方法选取健康雄性SD大鼠120只,随机分为假手术组、模型组、七叶皂苷钠组(简称治疗组)3组。每组又分为1、2、3和7 d 4个时间点,每个时间点5只大鼠,采用大鼠尾状核注入自体不凝血制备脑出血模型,大鼠分别在不同时间点断头取脑,采用Western-blot免疫印迹技术观察NF-κB蛋白表达,免疫组化染色观察AQP-4的变化。结果治疗组大鼠脑内NF-κB[(0.293±0.067),(0.441±0.064),(0.263±0.068),(0.258±0.061)]、AQP-4蛋白[(11.57±1.98),(14.27±2.16),(16.83±2.34),(9.82±1.64)]表达在不同时间点,与模型组比较差异均有统计学意义(P<0.05)。相关性分析两者呈正相关(r=0.82,P<0.05)。结论七叶皂苷钠可以抑制脑出血后NF-κB和AQP-4的表达,减轻脑损伤的程度。
Objective To study the dynamic expression of NF-κB and AQP-4 after intracerebral hemorrhage(ICH) rats with cerebral hemorrhage.Methods The 120 healthy SD male rats were randomly divided into 3 groups: sham group,model group,and treatment group,each group was examined at 4 different time points of 1 day,2 days,3 days and 7 days.ICH rat models were prepared with stereotactic infusion of autologous blood into caudate nucleus.The brains of rats were gotten off at different times,the expression of NF-κB were detected by Western-blot analysis and the expression of AQP-4 were detected respectively by immunohistochemistry staining.Results The expression of NFκB [(0.293±0.067),(0.441 ±0.064),(0.263 ±0.068),(0.258 ±0.061)] and AQP-4 [(11.57 ±1.98),(14.27 ±2.16),(16.83 ± 2.34),(9.82±1.64)] in the brain of the treatment group rats,showed significant differences compared with the model group(P &lt; 0.05).The correlation analysis was positively relative(r = 0.82,P &lt; 0.05).Conclusion Sodium aescinate can reduce the expression of NF-κB and AQP-4 after ICH and relieve the severity of brain damage.
