摘要
本文采用反相高效液相色谱建立了长柄扁桃仁系列产品中苦杏仁苷含量的测定方法。长柄扁桃仁系列产品中的苦杏仁苷经甲醇提取,以V(甲醇)∶V(水)∶V(乙腈)=2∶85∶13为流动相进行洗脱,在220nm检测波长下,测得长柄扁桃仁中苦杏仁苷的平均质量分数为1.44%。方法线性范围为50~300μg/mL,线性相关系数r=0.9996,检出限为1.2μg/mL,平均回收率为98.97%(n=5)。
A method of reversed-phase high performance liquid chromatography(RP-HPLC)has been developed for the determination of amygdalin in amygdalus pedunculatus pall products.The sample of amygdalin in amygdalus pedunculatus pall was extracted in methanol.The separation was perfomed with a mobile phase of methanol and water and acetonitrile(2∶85∶13)and a detection wavelength of 220nm.The average content of amygdalin in amygdalus pedunculatus pall detected by this method is 1.44%.The linear range was from 50to 300μg/mL(r=0.9996).The limit of detection for amygdalin was 1.2μg/ mL,the average recovery was 98.9%(n=5).The method is accurate,reproducible and can be used for the quality control of amygdalin.
