过表达GCNF对HeLa细胞生物学行为的影响

摘要 目的:将成功构建的人pcDNA3.1-GCNF真核表达载体瞬时转染宫颈癌HeLa细胞,观察GCNF基因对HeLa细胞生物学功能的影响。方法:将重组质粒pcDNA3.1-GCNF转染HeLa细胞,通过MTT比色法、基质胶黏附实验和Transwell侵袭试验对转染后HeLa细胞增殖、黏附和侵袭力进行分析。结果:成功获得瞬时转染的pcDNA3.1-GCNF HeLa细胞,转染目的基因组(0.239±0.014)与转染空质粒组(0.198±0.020)及未转染组(0.204±0.012)相比,细胞增殖最快(P<0.05)。转染目的基因后细胞粘附、侵袭力未见明显改变。结论:GCNF基因促进了体外培养的宫颈癌HeLa细胞增殖,但对HeLa细胞粘附、侵袭力没有明显的影响。 Objective: To transfect cervical cancer HeLa cells with constructed successfully human pcDNA3.1 -GCNF eukaryotic expression vector, and observe the effect of GCNF gene on biological behaviors of HeLa cells. Methods: HeLa cells were transfected with recombinant pcDNA3.1 - GCNF plasmid ; MTT colorimetric method, matrigel adhesion experiment, and Transwell invasive test were used to analyze proliferation, adhesion, and invasion abilities of HeLa cells after transfection. Results: HeLa cells after transient transfection with pcDNA3.1 - GCNF plasmid were obtained successfully, the cell proliferation speeds in pcDNA3.1 - GCNF plasmid transfection group, null vector transfection group, and non - transfection group were (0. 239 ±0. 014) , (0. 198±0. 020) , and (0. 204 ±0. 012) , respectively, the cell proliferation speed in pcDNA3. 1 - GCNF plasmid transfeetion group was the fastest ( P 〈 0. 05 ) ; after transfection, the adhesion and invasion abilities of HeLa cells didnt change significantly. Conclusion: GCNF gene promotes the proliferation of cervical cancer HeLa cells in vitro, but it has no impact on adhesion and invasion abilities of HeLa cells.
