缺氧诱导因子和一氧化氮合酶在肺栓塞大鼠肺组织中的表达及相关性研究 被引量:1

摘要 目的观察大鼠肺血栓栓塞症中肺组织中缺氧诱导因子和一氧化氮合酶的表达及血清NO水平的变化,探讨缺氧诱导因子和一氧化氮合酶对大鼠肺血栓栓塞症的影响及其作用机制。方法采用自体血栓回输法建立动物模型。随机分为对照组、PTE模型组,取动脉血测血气,常规病理学检测,用硝酸还原酶法检测血清中NO的水平,Western blotting和RT-PCR分别检测HIF-1、iNOS的表达。结果 PTE模型组肺组织病理损伤明显,各个时间点HIF-1及iNOSmRNA及蛋白水平较同时点正常组明显增高(P<0.05),且HIF-1及iNOS两者在mRNA及蛋白水平的表达均存在正相关(r=0.873、0.917,P<0.05)。结论 HIF-1及iNOS表达上调可能参与肺栓塞后机体代偿低氧的机制。 Objective To observe the change of expression of HIF - 1 and iNOS in acute pulmonary thromboembolism and evaluate their correlation. Methods Rats were randomly divided into control group, PTE model group. Rat PTE model was established by intravenous injection autologous blood clots. All rats were sacrificed on 1 hour,3 hour, lday, 3day,5day and 7day. I ml vascular blood was withdrawn from each animal to measure blood gas analysis. The pathological changes of lung were examined with light microscope. The expression of HIF - 1 and iNOS were evaluated by RT - PCR and Western blotting method. The plasma of NO were measured by biochemisty. Results FFE model group lung his- topathologic injury was showed evidently; Compared with control group, the expression of HIF- 1 and iNOS mRNA and protein obviously increased in pet group at every time length( P 〈 0. 05 ). The level of NO begun to elevated at 1 hour af- ter embolization. Conclusions the up - regulated expression of HIF - 1 and iNOS after the PTE perhaps participate in the compensated mechanism of hypoxia, thromblytic and safflor.
