摘要
目的通过检测转录因子KLF5蛋白和细胞周期素D1(Cyclin D1)、增殖细胞核抗原(PCNA)、P53蛋白在结肠癌组织中的表达,对KLF5在结肠癌细胞增殖过程中的意义进行探讨。方法对48例结肠癌和30例癌旁组织石蜡标本进行免疫组化染色,检测KLF5与Cyclin D1、PCNA、P53在两种组织中的表达情况,并分析其关系。结果结肠癌组织中KLF5、Cyclin D1、PCNA表达均强于癌旁组织(均P<0.05),P53在两种组织中表达无明显差异(P>0.05)。相关分析显示,结肠癌组织中KLF5与Cyclin D1之间、KLF5与P53之间表达存在正相关关系(P均<0.05);Cyclin D1与PCNA表达也存在正相关关系(P<0.05),Cyclin D1与P53、PCNA与P53之间表达均无明显相关关系(P>0.05)。结论结肠癌组织中KLF5表达上调,这与KLF5可能通过调节CyclinD1、P53表达而参与结肠癌细胞增殖有关。
Objective To nvestigate the significance of the expressions of Kruppel-like factors 5 (KLFS) ,Cyclin D1, proliferating cell nuclear antigen (PCNA), P53 during cell proliferation in colon cancer. Methods The expression levels of KEFS, Cyclin D1, PCNA, P53 were detected by immunohistochemistry in 48 cases of colon cancer paraffin-embedded specimens and 30 cases of carcinoma-adjacent tissues, and the correlation among them was analyzed. Results The expression levels of KLF5 ,Cyclin D1 ,PCNA in colon cancer tissues were significantly higher than those in carcinoma-adjacent tissues ( P 〈 0. 05 ), howecer, there were no significant differences in the expression levels of P53 between the two kinds of tissues ( P 〉 0. 05). The results of correlation analysis showed that KLF5 was positively correlated to Cyclin D1, so to P53 ( P 〈 0. 05) ;Cyclin Dlexpression was positively related to that of PCNA ( P 〈0.05 ) ,however,there was no correlation between Cyclin D1 and P53 and between PCNA and P53 ( P 〉 0. 05 ). Conclusion The expression levels of KLF5 are up-regulated in colon cancer tissues ,which may be involved in the cell proliferation of colon cancer by regulating the expression levels of Cyclin D1 and P53.
