摘要
目的建立高效液相色谱法实时监测患者血浆中甲氨蝶呤浓度,为临床合理用药提供参考。方法以Agilent Eclipse XDB-C18柱(4.6×150mm,5μm)为色谱柱,流动相为醋酸(pH=2.86)-甲醇(82.5:17.5,v/v),流速:0.8ml/min,检测波长306nm,柱温柱温:40℃,样品经高氯酸沉淀蛋白后直接进样。结果表明血浆中甲氨蝶呤的质量0.3~10.20μg/ml内线性关系良好,r=1(n=3),检测限为0.03μg/ml。方法回收率为96.43%~99.49%,提取回收率为:77.04%~85.61%。日内和日间RSD均<5%。结论该方法简便、准确、重复性好,适用于甲氨蝶呤血药浓度监测。
Objective To establish a HPLC method for the determination of the plasma concentration of methotrexate(MTX).Methods Seperation and determination were achieved on a Agilent Eclipse XDB-C18 column(4.6 × 150 mm,5μm)with the mobile phase consisted of acetic acid(pH = 2.86)methanol(82.5: 17.5)at a flow rate of 0.8 ml/min.The UV detection wavelength was set to 306nm.The column temperature was 40℃ .The plasma samples were treated with perchloric acid for sedimentation.Results The good linearity was from 0.3 to 10.20 μg/ml,r = 1(n = 3).The minimum detectable concentration for MTX was 0.03 μg/ml.The method recoveries of methotreexate in high,medium and low concentrations were from 96.43% to 99.49% ,and the extraction recoveries were from 77.04% to 85.61% .The within day and between day precision of variation was less than 5% .Conclusion This method is simple,accurate and reliable.It is suitable for the blood drug concentration monitoring of MTX in clinic.
