转SsBHMT基因拟南芥植株的鉴定及其耐缺硫性分析

摘要 硫是植物的必须营养成分之一,硫供应不足不仅影响到作物的产量而且影响到品质。本研究以转甜菜碱高半胱氨酸甲基转移酶基因(SsBHMT)拟南芥植株为材料,利用RT-PCR、Western blot等方法,鉴定了转基因植株,分析了其在阻断外界硫供应条件下的生理变化。结果表明:与野生型相比,缺硫胁迫条件下转基因拟南芥叶片枯黄程度低,具有较高的SOD活性、叶绿素含量,较低的MDA含量;SsBHMT基因的超表达提高了拟南芥的抗缺硫能力。 Sulfur is an essential macroelement in plant nutrition. A lack of sulfur supply affects harvest and nutritional quality of crop plants. In this study, SsBHMT transgenic Arabidopsis were identified by RT-PCR and western blot. The physiological traits of transgenic Arabidop- sis plants were analysed during sulfur limitation. The results showed that the transgenic plants exhibited less discoloration in comparison to control plants under sulfur deficiency stress. The activity of SOD and chlorophyll content were higher than wild type plants. The content of MDA was lower than wild-type. The overexpression of SsBHMT effectively increased the sulfur deficiency tolerance capacity in transgenic plants.
