新疆番茄黄化曲叶病毒检测及其DNA-A基因组序列分析 被引量:10

摘要 2012年,在新疆喀什、吐鲁番和伊犁地区发现番茄、辣椒、茄子、曼陀罗、仙客来、一品红和红掌等7种植物表现矮化、叶片黄化、卷曲等疑似双生病毒的感病症状。检测结果表明:以上7种植物均能被番茄黄化曲叶病毒(To-mato yellow leaf curl virus,TYLCV)侵染。本试验共获得8个TYLCV病毒分离物,分别为番茄分离物KSCTo4、KSCTo16、KZPTo18、KYCTo20,辣椒分离物KSCPe6、KSCPe7,茄子分离物KSCEg1,曼陀罗分离物KSCDa。8个分离物全部序列均具有菜豆金色花叶病毒属Begomovirus病毒基因组典型特征,分离物在不同寄主上的序列差异较小,序列间相似性达99.0%~99.8%,与中国已报道的TYLCV分离物SH3、ZJ6、SD2A-7、HNSQ、HBBD的相似性达98.6%~99.5%。 In 2012, yellow leaf curl symptoms were observed in tomato, pepper, eggplant, datura, poinsettia, cyclamen and anthurium in Kashi, Turfan and Yili prefectures of Xinjiang. The results showed that all of the above 7 species of plants could be infected by Tomato yellow leaf curl virus (TYLCV). The full-length DNA-A ge- nomic sequences of the isolates KSCTo4, KSCTo16, KZPTol8 and KYCTo2O from tomato, KSCPe6 and KSCPe7 from pepper, and KSCEgl from eggplant and KSCDa from datura were obtained. Each isolate had all of the characteristic fea- tures of Begomovirus genome organization. There were no big differences in DNA sequence of the isolates obtained in this study between different hosts. Pairwise sequence comparisons of the 8 isolates indicated that the identities were 99. 0% --99.8%. And these DNA-A sequences had higher identity with the reported isolates SH3, ZJ6, SD2A-7, HNSQ and HBBD of TYLCV from other regions of China, with a sequence identities of 98.6%--99.5%.
