摘要
目的:探讨血府逐瘀汤对大鼠局灶性脑缺血模型的保护作用。方法:采用线栓法制作大鼠大脑中动脉栓塞模型(MCAO)。将60只健康成年雄性SD大鼠按随机数字表法分为空白对照组、模型对照组、阳性对照组、血府逐瘀汤低剂量组、中剂量组、高剂量组,每组各10只。测定脑组织中的谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)含量、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及一氧化氮合酶(NOS)活性,并进行行为学评分、血液流变学和病理组织学检查。结果:大鼠的行为学表现,造模后的各组大鼠均出现明显的行为学障碍,血府逐瘀汤治疗组大鼠行为学表现均有缓解的趋势;阳性对照组和血府逐瘀汤高剂量组的大鼠行为学障碍得到明显改善,且与模型组比较,差异有统计学意义(P<0.05);与空白对照组相比,模型对照组的全血黏度、血浆黏度及红细胞比容都显著升高(P<0.05)。与模型对照组比较,各给药组全血黏度、血浆黏度显著降低(P<0.05);阳性对照组和血府逐瘀汤高剂量组的红细胞比容显著降低(P<0.05)。与空白对照组比较,模型对照组大鼠脑组织中的MDA含量、NOS活性明显升高,SOD、GSH-Px、CAT活性明显降低(P<0.05);与模型对照组比较,血府逐瘀汤治疗组可降低缺血脑组织MDA含量和NOS活性,提高缺血脑组织中SOD、GSH-Px、CAT的活性(P<0.05)。结论:血府逐瘀汤能减缓MCAO模型的行为学障碍,明显提高缺血脑组织中SOD、GSH-Px、CAT活性,降低MDA含量,并有降低缺血脑组织中NOS活性的趋势。血府逐瘀汤对局灶性脑缺血模型有一定的保护作用。
Objective:To study the protective effect of Xuefu Zhuyu decoction for focal cerebral ischemia.Methods:Use line to establish middle cerebral artery occlusion model rats.Healthy adult male SD rats were randomly divided into blank control group,model control group,positive control group and therapeutic low dose group,middle dose group and high dose group,10 in each group.Determine the brain glutathione peroxidase(gsh-px),malondialdehyde(MDA) content,superoxide dismutase(SOD),catalase(CAT) and nitric oxide synthase(NOS) activity as well as behavioral score,blood rheology and histopathologic examination.Results:Each group of model rats showed obvious behavioral disorder.The behavior performance of therapeutic treatment group rats has a tendency to ease.The behavioral disorder of positive control group and therapeutic high dose group of rats had dramatically improved.The difference was statistically significant compared with model group(P0.05).Compared with the blank control group,the whole blood viscosity,plasma viscosity and erythrocyte volume of model control group were significantly higher(P0.05).Compared with model control group,the whole blood viscosity and plasma viscosity of each dose group were decreased significantly(P0.05).The red blood cell volume of positive control group and therapeutic high dose group was decreased significantly(P0.05).Compared with blank control group,the MDA content and NOS activity of model control group were increased significantly but SOD,GSH-Px and CAT activity were decreased significantly(P0.05).Compared with model control group,therapeutic treatment group can reduce MDA content and NOS activity in ischemic brain tissue as well as improve the SOD,GSH-Px and CAT activity(P0.05).Conclusion:Xuefu Zhuyu Decoction can slow down the behavioral disorders of MCAO model,obviously improve the SOD,GSH-Px and CAT activity in ischemic brain tissue and decrease the MDA content as well as a tendency to reduce the NOS activity in ischemia brain tissue.Xuefu Zhuyu Decoction has certain protective effect on focal cerebral ischemia model.
