摘要
目的:研究六味地黄丸联合山楂对高脂饮食诱导的非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)模型大鼠的防治作用。方法:在50只健康SD大鼠中随机抽取10只为正常对照组(雌雄各半),其条40只运用高脂饲料喂养12周诱导NAFLD模型,造模成功后将其随机分为模型组、中药低剂量组、中药高剂量组及西药对照组各10只,雌雄各半。自造模成功之日起对各组大鼠进行灌胃干预。中药低、高剂量组分别用六味地黄丸联合山楂浓缩水煎液灌胃,剂量分别为1.2mL/100g体质量(浓度为0.48g/mL)、2.4mL/100g体质量(浓度为0.96g/mL);西药对照组用非诺贝特混悬液灌胃,剂量为1mL/100g体质量(浓度为4mg/mL);模型组给予等体积蒸馏水灌胃,均每天1次,持续2周。灌胃结束后,观察各组药物对大鼠血清肝功能、血脂和肝脏组织病理变化的影响。结果:与模型组相比,中药低、高剂量组大鼠血清谷丙转氨酶(ALT)、血清总胆固醇(TC)显著降低(P<0.05),中药高剂量组血清三酰甘油(TG)显著降低(P<0.05),肝细胞脂肪空泡数量明显减少,肝组织脂肪变性明显改善。结论:六味地黄丸联合山楂煎剂可显著降低高脂饮食诱导的大鼠NAFLD模型中ALT和TC含量,减轻肝脂肪变性,对NAFLD动物模型有一定疗效。
Objective :To investigate the effect of Liuwei Dihuang pills combined with hawthorn decoction in the prevention and treatment of non- alcoholic fatty liver disease (NAFLD) induced by high -fat diet among rats. Methods:In 50 healthy SD rats were randomly selected 10 rats into normal group( male and female) ,the remaining. Forty rats were fed with high -fat feed for 12 weeks to establish a rat model of NAFLD. Experimental rats were randomly and equally divided into, model group ( n = 10), low - dose traditional Chinese medicine (TCM) group ( n = 10), high - dose TCM group ( n = 10), and Western medicine control group ( n = 10), ( male and femde). After the rat model of NAFLD was established, the low - dose and high - dose TCM groups received Liuwei Dihuang pills com- bined with concentrated water decoction of hawthorn fruit (0. 48 g/ml, 1.2 ml/100g body weight ; 0. 96 g/ml, 2.4 ml/100g body weight) by gavage, the Western medicine control group received fenofibrate suspension (4 mg/ml, 1 ml/100g body weight) by gavage, and the model group received an equal volume of distilled water. Intragastric administration was performed once daily for 2 weeks. The therapeutic effects were assessed based on the liver function indices and blood lipid levels in serum and the pathological changes of liver tissue. Results : Compared with the model group, the low - dose and high - dose TCM groups had significantly decreased serum alanine aminotransferase (ALT) and total cholesterol (TC) levels (P 〈 0. 05 ), and the high -dose TCM group had a significantly decreased serum triglyceride level (P 〈 0. 05), as well as significantly decreased number of fat vacuoles in hepatocytes and significantly reduced fatty degeneration of liver tissue. Conclusion:Liuwei Dihuang pills combined with hawthorn decoction can significantly decrease serum ALT and TC levels and significantly reduce fatty degeneration of liver tissue in the rat model of NAFLD induced by high - fat diet and have certain therapeutic efficacy for the animal model of NAFLD.
