摘要
目的探讨子宫内膜组织中端粒酶催化亚单位(human telomerase reverse transcriptase,hTRT)表达与DNA含量对子宫内膜癌的诊断意义以及二者之间的关系。方法采用原位杂交法对子宫内膜癌40例、子宫内膜不典型增生20例及正常子宫内膜组织8例的hTRT表达进行检测;用流式细胞仪检测3种组织细胞的DNA含量。结果子宫内膜癌组织hTRT表达阳性率为87.5%,明显高于正常子宫内膜与子宫内膜不典型增生组织(P<0.01);而且子宫内膜不典型增生组织hTRT表达阳性率(40.0%)高于正常子宫内膜(12.5%)。从正常子宫内膜到子宫内膜不典型增生及子宫内膜癌,DNA指数值与S期细胞百分比值逐渐增高(P<0.05)。hTRT阳性肿瘤组织中异倍体肿瘤(28/35)明显高于hTRT阴性肿瘤组织(1/5)。结论从正常子宫内膜到子宫内膜不典型增生及子宫内膜癌,hTRT表达阳性率及DNA含量逐渐增高,提示hTRT阳性表达和DNA含量的增高与子宫内膜癌的发生发展高度相关,对子宫内膜癌的诊断具有重要意义。
Objective To investigate the value of human telomerase reverse transcriptase (hTRT) and DNA contents in the diagnosis of endometrial carcinoma and to evaluate the relationship between the expression of hTRT and DNA content in endometrial carcinoma. Methods The expression of hTRT in 40 cases of endometrial carcinoma, 20 cases of endometrial atypical hyperplasia and 8 cases of normal endometrium were detected by in situ hybridization. The DNA contents of the three kinds of tissue were detected by flow cytometry. Results The positive rate of hTRT in endometrial carcinoma (87.5 % ) was remarkably higher than those in endometrial atypical hyperplasia (40.0%) and normal endometrium (12.5%) ,respectively(P 〈0.01 ). The difference of DNA index and S-phase fraction among the three groups were significant ( P 〈 0. 05 ). The aneuploid rate ( 28/35 ) of hTRT-positive carcinoma was significantly higher than that ( 1/5 ) of hTRT-negative ( P 〈 0. 05 ). Conclusion From normal endometrium to endometrial atypical hyperplasia and endometrial carcinoma, the expression of hTRT and DNA content increased gradually, indicating hTRT positive expression and increased DNA content being closely related to the progress of endometrial carcinoma, both of them played important role in the diagnosis of endometrial carcinoma.
