摘要
作者用碳二亚胺及二元胺使羧化乳胶转变为胺化乳胶,再以异刷双功能剂3-(2—吡啶二硫)丙酸琥珀酰亚胺酯(SPDP)引入二硫吡啶基(PDP),得PDP化乳胶;后者与引入巯基的牛血清白蛋白(BSA)反应而使乳胶致敏.结果显示,此法致敏的乳胶在凝集试验中具有较高的敏感性和特异性,效价可达到1:7680,用抗人血清白蛋白或正常兔血清代替抗BSA血清,均未见凝集现象.
A highly sensitive and specific immunolatex was prepared first by conversion of carboxylated latexes into aminated latexes with carbondiimide and diamine, then introduction of 2 - pyridyldithio groups (PDP) with N - succinimidyl 3 - (2 - pyridyldithio ) propionate (SPDP ) and finally by the reaction of the resultants with a protein used as antigen into which hydrosulfuryl groups have been induced already . The latex agglutination test shows that the sensitized latexes obtained have a valence of 1 : 7 680, about 25 times as high as the conventional me -thods.None agglutination was observed when an anti - HSA or normal rabbit scrum was substituted for the anti- BSA scrum .
