大鼠C6脑胶质瘤晚期阶段模型MR成像及弥散张量研究 被引量:1

摘要 目的探讨3.0T磁共振成像及弥散张量成像技术对大鼠C6脑胶质瘤模型的应用价值。方法大鼠C6脑胶质瘤细胞体外培养,通过立体定向技术将C6细胞接种至30只雌性Wistar大鼠右侧尾状核。接种3周左右,应用3.0T高场强医用磁共振扫描仪对大鼠行常规MRI、DTI及增强T1WI检查,并与病理结果对照。结果 30只接种大鼠中,28只在随后的MRI和病理检查中均证实有肿瘤形成,成瘤率约93%。磁共振成像呈长T1长T2信号,信号欠均匀,增强扫描呈明显均匀强化或环状强化。DTI显示瘤体和对侧镜像脑组织平均FA值分别为(0.12±0.04)和(0.31±0.05),而平均MD值分别为(0.96±0.04)×10-3mm2/s和(0.79±0.02)×10-3mm2/s,差别均有统计学意义(P<0.05)。结论 Wistar大鼠C6脑胶质瘤模型稳定可靠,接种成功率高。用临床3.0T MRI行大鼠C6胶质瘤DTI检查切实可行,通过测量瘤体及对侧镜像脑组织FA及MD值,为以后的科研工作提供了有用的参考。 【Objective】 To explore the application value of magnetic resonance imaging and diffusion tensor imaging in the late stage of wistar rat C6 brain glioma.【Methods】 Rat C6 glioma cells were cultured in vitro and the concentration of more than 1.0 ×106/10 μL glioma cells and complete medium were injected stereotactically into the right caudate nucleus of 30 Wistar rats.Conventional MRI,DTI,and enhanced T1WI and T1-3D-IR-FSPGR scans were performed using the GE Signal HDx 3.0T MRI scanner about 3 weeks after implantation for the rats.【Results】 28 tumor bearing rats were confirmed the tumor formation by the subsequent MRI and pathological examination,with a success rate of 93%.MRI scans showed the rat C6 glioma were long T1,long T2 signal,homogeneous enhancement or ring enhancement after injection of contrast.T1-3D-IR-FSPGR series three-dimensionally showed tumor size,and adjacency and internal structure etc.The average FA values of 3 equal-sized ROIs in the 28 wistar rats C6 brain glioma regions and the contralateral normal brain parenchyma were(0.12 ± 0.04) and(0.31 ± 0.05),respectively;The average MD values were(0.96 ± 0.02) ×10-3 mm2/s and(0.79 ± 0.05) ×10-3 mm2/s,respectively.Two independent-sample T test of the two groups showed statistically significant difference(P 0.05).【Conclusion】 Wistar rat C6 glioma mod-el is a stable and reliable model.After implantation,tumor grows rapidly,and the success rate is very high.It is feasible to perform DTI scan in the rat C6 brain glioma model using a clinical 3.0T MR scanner,and the FA and MD values measured in the rat C6 brain glioma regions and the contralateral normal brain parenchyma can be used for reference in future studies.
