摘要
报道了二芳基甲基哌嗪阿片受体激动剂DPI-3290的一种新的合成路线.该合成的关键步骤是一个四组分三步串联反应,通过对这一反应的条件和操作的优化,以41%的较好产率得到了预期产物.应用这一路线,以四步直线步骤和16.8%的总产率实现了DPI-3290的克级制备.
A practical synthesis of benzhydrylpiperazine opioid receptor agonist DPI - 3290 was developed. The key step in this synthesis was a 4 - component 3 - stage cascade reaction. After the optimization of the reaction condi-tions and operation, the expected condensation product was obtained with a 41% yield, synthesis of DPI-3290 proved to be effective in scale-up trials, affording about 16.8% overall yield in 4 steps.
