摘要
目的获得深圳口岸首例入境人员HIV-1亚型毒株全长基因组序列并分析其特征。方法以深圳出入境检验检疫局HIV确证实验室确证的1例HIV感染者为研究对象,提取病毒RNA,利用逆转录巢式RT-PCR技术,分两段扩增病毒近似全长基因序列,对PCR产物进行核苷酸序列测定,应用生物信息学软件对全基因序列进行系统进化分析。结果获得了8 831 bp长度的近全长基因组序列,系统进化树分析结果显示该毒株与HIV-1 CRF01_AE亚型参考株成簇,Bootstrap值为100%,通过与世界上不同国家和地区流行的HIV-1 CRF01_AE亚型毒株比较,该毒株与泰国流行毒株的关系最近。结论该毒株是由境外传入的,应对入境人员HIV感染毒株的来源和变异进行密切监测。
Objective To gain the full-length genome sequence of the first HIV-1 virus strain among entry personnel at Shenzhen port, and to analyze its characterization. Methods Peripherial blood plasma was collected. The viral genomic RNA was extracted and amplified in two halves by using nested RT-PCR. The PCR products were sequenced after being purified. The sequence was analyzed by biological infatuation software. Results The length genomic sequence of 8831 bp was acquired. The results of analysis of phylogenetic relationship suggested the strain was subtype CRF01_AE, which was further proved by the phylogenetie tree with a high bootstrap vlue (100%). In the phylogenetic tree, the isolated strain was group into clusters with those from. Conclusion The strain was introduced from abroad. It is necessary to enhanced survillance for HIV-1 among entry-enxit personnel.
