京尼平-1-β-D-龙胆双糖苷对大鼠心力衰竭的影响 被引量:2

The cardiac systolic and diastolic effects of genipin-1-β-D-gentiobioside in the experimental heart failure

摘要 目的:研究京尼平-1-β-D-龙胆双糖苷(genipin-1-β-D-gentiobioside,GG)对在体心力衰竭模型大鼠及正常离体大鼠心脏收缩和舒张功能的影响。方法:在体心力衰竭模型大鼠实验:采用左心室插管法测定大鼠心率(HR)、左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室内压最大上升速率(+dp/dt max)和左室内压最大下降速率(-dp/dt max)。正常离体大鼠实验:用Langendorff心脏灌注装置记录离体心脏的HR、LVSP、LVEDP、dp/dt max和-dp/dt max。结果:戊巴比妥钠可诱发大鼠心力衰竭,GG(40 mg/kg)使减慢的HR明显加快,降低的LVSP和dp/dt max增加,升高的LVEDP降低。在离体心脏实验中,GG(1,10,100μmol/L)浓度依赖性增加LVSP和+dp/dt max,降低LVEDP,但对HR和-dp/dt max无明显影响。结论:GG通过提高心脏的收缩功能及降低其前负荷,改善戊巴比妥钠引起的心力衰竭。 This study was to investigate the cardiac systolic and diastolic effects of Genipin-l-β-D-gentiobioside (GG) in the experi- mental heart failure and normal rats. Methods: in vivo experimental heart failure model experiments: the heart rate (HR), the peak systolic pressure ( LVSP), left ventrlcular end-diastolic pressure ( LVEDP), peak rate of rise of left ventricular pressure ( + dp/dt max), and peak rate of fall of left ventricular pressure ( -dp/dt max) were measured by a polyethylene tube inserted into the left ventricle via the right carotid artery, in vitro normal isolated rat heart experiments: the HR, LVSP, LVEDP and + dp/dt max were measured by the conventional Lange- ndroff method. Results: GG (40 mg/kg) significantly decreased the LVEDP and increased the HR, LVSP, + dp/dtmax and -dp/dt max in heart failure rats induced by pentobarbital sodium. Moreover, GG (1, 10, 100 μmol/L) significantly enhanced the LVSP and + dp/dt max and decreased the LVEDP in a concentration dependent manner without remarkably affecting the HR and -dp/dt max. Conclusion: Effects of Genipin-1-β-D-gentiobioside on protecting against heart failure of rats induced by pentobarbital sodium were mediated by enhancing rat heart inotropy and lowering the preload of heart.
