摘要
目的 :探讨黄芪多糖(Astragalus polysaccharides,APS)对卡介苗(BCG)和脂多糖(LPS)诱导肝损伤的保护作用及其可能的机制。方法:60只SD大鼠随机分为正常组;BCG+LPS模型组;黄芪多糖(200、400、800 mg/kg)治疗组和联苯双酯(150 mg/kg)对照组。治疗组和对照组给予相应APS或联苯双酯连续灌胃10天,每日一次。模型组、治疗组以及对照组尾静脉注射BCG和LPS,正常组尾静脉注射等体积的生理盐水。尾静脉注射LPS 12小时后,摘眼球取血,比色分析法测定小鼠血清谷丙转氨酶(AST)、谷草转氨酶(AL T)的活性;HE染色观察肝组织形态学变化;称重法测定肝脏指数、脾脏指数;以ELISA法测定血清中TNF-α,IL-6水平;Western blot测定肝组织中p65,IκBα表达。结果:与正常对照组相比,模型组中A S T、AL T活性增高;肝脏指数、脾脏指数增高;TNF-α,IL-6含量增加;p65表达增高而IκBα表达降低。黄芪多糖(200、400、800 mg/kg)在降低A S T、AL T活性的同时,可降低BCG+LPS诱导的肝脏指数、脾脏指数的增加,使TNF-α,IL-6水平降低,p65表达降低而IκBα表达增高。且APS对肝损伤的这种保护作用呈现一定的剂量依赖性。结论:黄芪多糖对免疫性肝损伤具有保护作用,且这种保护作用可能与黄芪多糖降低炎症因子水平,抑制NF-κB通路有关。
The aim of the present study was to study the protective effect of Astragalus polysaccharides on immunological liver injury in- duced by BCG and LPS,and to explore a possible mechanism. Methods: 60 SD rats were randomly divided into 6 groups: the normal group, the model group, the different does of Astragalus polysaccharides groups (200,400,800 mg/kg), and the positive control biphenyl group (150 mg/kg) ,with 10 mice in each group. The treatment group and positive control group were given Astragalus polysaccharides or bifendate daily for 10 days. Immunological liver injury in mice was induced by BCG and LPS, after administration of LPS for 12 hours, the rats were killed and the activity of serum ALT, AST, the liver index and spleen index were detected. Liver pathological changes were observed by HE stai- ning. The content of TNF-ct and IL-6 were determined by ELISA; the expression of p65 and IKBot were measured by Western blot. Results: compared with the normal group, the activity of ALT,AST, the liver index and spleen index, the content of TNF-et and IL-6 and the expres- sion of p65 were increased while the expression of IKBcx was decreased. APS(200,400,800 mg/kg) could obviously reduce the activities of ALT,AST, at the same time reduced the liver index and spleen index, the content of TNF-α and IL-6 and the expression of p65, increased the the expression of IKBα compared with model group. Conclusion: APS has a protective effect on immunological liver injury, which is par- tially via attenuating inflammatory effect and the NF-KB signling pathway.
