摘要
目的:通过高脂饮食诱导大鼠胰岛素抵抗(insulin resistance,IR)模型,观察抵当汤对其主动脉组织中胰岛素受体(insulin receptor,InsR)及胰岛素样生长因子1(insulin-like growth factor 1,IGF-1)以及血清中IGF-1的影响,探讨抵当汤改善胰岛素抵抗的作用机制。方法:雄性大鼠35只,随机分为5组,分别为:空白对照组、模型组、抵当汤低剂量组、抵当汤高剂量组、血脂康组。空白对照组喂普通饲料;模型组喂高脂饲料;抵当汤低、高剂量组喂高脂饲料,同时灌胃抵当汤煎剂1g/kg、2g/kg;血脂康组喂高脂饲料,同时灌胃血脂康0.2 g/kg,连续喂养8周。测定血清空腹血糖(FBG)、空腹胰岛素(FINS)和IGF-1,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR);胸主动脉应用免疫组织化学方法,观察主动脉组织中InsR和IGF-1的变化。结果:抵当汤低、高剂量组与模型组相比,大鼠HOMA-IR明显降低,ISI明显升高(P<0.05或P<0.01);抵当汤低、高剂量组与模型组相比,大鼠平滑肌细胞胞膜及胞浆InsR表达较强,平均光密度(average optical density,AOD)统计学有显著性差异;抵当汤低、高剂量组与模型组相比,大鼠血清IGF-1明显升高(P<0.05);抵当汤低、高剂量组与模型组相比,大鼠平滑肌胞膜及胞浆IGF-1表达明显较强,AOD统计学具有显著性差异(P<0.05)。结论:抵当汤可降低IR大鼠HOMA-IR,升高ISI,提高胰岛素敏感性;增强IR大鼠平滑肌胞膜及胞浆中InsR表达,升高IR大鼠血清及主动脉组织中IGF-1水平,改善胰岛素抵抗。
To observe the effect of Didangtang on the levels of insR and IGF-1 in insulin resistance rats. Methods:35 health and clean level Wistar male rats were chosen and randomly divided into 5 groups : control group, model group, low- dose Didangtang group, high-dose Didangtang group and Xuezhikang group with 7 rats each. The rats of control group were fed with normal diet and distilled water ( 10ml/kg), model group was fed high fat diet and distilled water ( 10ml/kg), low- dose Didangtang group was fed high fat diet and Didingtang ( 1g/kg), high-dose Didangtang group was fed high fat diet and Didingtang(2g/kg),Xuezhikang group was fed high fat diet and Xuezhikang(0.2g/ kg). After 8 weeks of feeding, take blood by decapitate. Serum FBG, FINS and IGF-1 were detected, HOMA-IR and ISI were calculated. Thoracic aorta was retained. Immunohistechemistry was used to observe the InsR and IGF-1 changes of aortic tissue. Results: Didangtang can markedly reduce the IR rats HOMA-IR, increased ISI; improve IR rats of insulin receptor expression. Increase IR rats in serum and aortic tissue IGF-1 levels. Conclusion: Didangtang can improve insulin sensitivity. Didangtang is an ideal medicine in preventing and treating dyslipidemia.
