摘要
[目的]探讨独活寄生汤对类风湿关节炎(RA)模型大鼠骨损伤的保护作用及其机制。[方法]建立II型胶原蛋白诱导的SD大鼠关节炎模型。将60只SD大鼠随机分成4组:空白组、模型组、来氟米特片组和独活寄生汤颗粒剂组,分别给予相应的药液灌胃,持续6周。采用原位杂交法测定大鼠血清中OPG、RANKL的表达水平。[结果]模型组与空白组相比,血清RANKL的平均光密度值明显升高,血清OPG的平均光密度值则明显降低(P<0.05)。[结论]独活寄生汤配方颗粒剂通过上调模型鼠血清中OPG表达,提高OPG/RANKL的比例,竞争性抑制RANK和RANKL的结合,从而有效延缓RA的骨侵蚀。
[Objective]Investigate Duhuojisheng Decoction on rheumatoid arthritis(RA) protective effect and mechanism of bone injury model of rats.[Methods]Establishment of type II collagen induced arthritis rat model of SD.60 SD rats were randomly divided into 4 groups: blank group,model group,leflunomide tablets group and Duhuo Jisheng Decoction granules group,were given by gavage for 6 weeks.To measure the expression of OPG,RANKL in serum of rats by in situ hybridization.[Results]The model group compared with the control group,the average optical density and the value of serum RANKL increased significantly,the average optical density value of the serum OPG was decreased significantly(P〈0.05).[Conclusion] Duhuojisheng Decoction granules through the expression of serum OPG increased in rat model,to improve the ratio of OPG / RANKL,combined with the competitive inhibition of RANK and RANKL,thus effectively delay the RA bone erosion.
