摘要
目的探讨蕨麻正丁醇(Pa)提取物对心肌缺血再灌注损伤大鼠白细胞介素-8(IL-8)的影响。方法Wistar雄性大鼠40只,采用随机数字表法分为对照组、缺血再灌注模型组和Pa提取物高、中、低剂量预处理组,每组8只,右心房取血酶联免疫吸附测定法检测血清IL-8,反转录-聚合酶链反应、Western blot技术检测心肌组织中IL-8mRNA及蛋白水平的表达。结果与对照组比较,模型组和Pa提取物中、低剂量预处理组血清IL-8水平及心肌组织IL-8 mRNA和蛋白表达水平均明显升高(P<0.05);Pa提取物高剂量预处理组血清IL-8水平及心肌组织IL-8 mRNA和蛋白表达水平与对照组比较差异无统计学意义(P>0.05);与模型组比较,Pa提取物高、中、低剂量预处理组血清IL-8水平及心肌组织IL-8 mRNA和蛋白表达水平明显下调(P<0.05);Pa提取物高剂量预处理组血清IL-8水平及心肌组织IL-8 mRNA和蛋白表达水平明显低于中、低剂量预处理组(P<0.05);Pa提取物中剂量预处理组血清IL-8水平及心肌组织IL-8 mRNA和蛋白表达水平明显低于低剂量预处理组(P<0.05)。结论 IL-8在大鼠心肌缺血再灌注损伤过程中表达上调,用Pa提取物进行干预可降低IL-8的表达。
Objective To investigate the effect of potentilla anserine (Pa) extractive on interleukin-8 ( IL-8 ) in acute myocardial ischemia-reperfusion(MI/R) injury rats. Methods Forty male Wistar rats were divided into control group, ische- mia-reperfusion model group, Pa high dose preconditioning group, Pa middle-dose preconditioning group and Pa low-dose pre- conditioning group by random digits table, with eight rats in each group. The serum IL-8 levels were determined by enzyme linked immunosorbent assay and the expression of IL-8 mRNA and protein level in myocardium tissue were determined by re- verse transcription-polymerase chain reaction and Western blot. Results The serum levels of IL-8,the IL-8 mRNA and protein expression in myocardium tissue in model group and Pa middle-dose preconditioning group and Pa low-dose preconditioning group were significantly higher than those in control group( P 〈 0.05 ) ;but there were not statistic differences between control group and Pa high-dose preconditioning group(P 〉 0. 05 ). Compared with model group, the serum levels of IL-8, IL-8 mRNA and protein expression in myocardium tissue were decreased in Pa high dose preconditioning group,Pa middle-dose preconditioning group and Pa low-dose preconditioning group(P 〈 0.05 ) ;and the serum levels of IL-8, IL-8 mRNA and protein expression in myoeardium tissue in Pa high dose preconditioning group were significantly lower than those in Pa middle-dose preconditioning group and Pa low-dose preconditioning group(P 〈 0.05 ) ;and the serum levels of IL-8,IL-8 mRNA and protein expression in myocardium tis- sue in Pa middle-dose preconditioning group were significantly lower than those in Pa low-dose preconditioning group (P 〈 0. 05). Conclusion IL-8 level increase in MI/R rats. Pa extractive can reduce the expression of IL-8.
