摘要
目的 为研究治疗白血病的有效药物。方法 将槲皮素作用于培养的 NB4细胞株 ,应用 RT- PCR及western blot方法 ,研究不同浓度槲皮素 (30、6 0、90μm)诱导细胞 2 4、48、72、96、12 0 h后 ,白血病 NB4细胞株中突变型 p5 3基因及蛋白表达的情况。结果 证实其 30~ 90 μm浓度时对突变型 p5 3基因没有影响 ,而对突变型 p5 3蛋白的表达产生抑制作用。
Objective In order to seek a valuable drug for the treatment of lukmia,Quercetin was tested in cultured NB4 cell line.Methods The expression of mutant p53 gene and protein after induction of Quercetin in different concentrations of 30,60,90 μm for 24,48,72,96 h and 120 h respectively was tested by using RT PCR and Western blot.Results Quercetin with concentrations of 30~90 μm could successfully inhibit the expression of mutant p53 protein,but not mutant p53 gene in NB4 cell line.Conclusion Quercetin seems to be an effective drug for the treatment of leukemia.
