摘要
目的:探讨Peroxiredoxin(PRDX) Ⅳ mRNA在小鼠卵泡发育和卵巢衰老过程中表达。方法:选用C57BL/6j小鼠不同周龄、动情周期以及促性腺激素预处理卵巢,通过实时定量聚合酶链反应检测PRDX Ⅳ mRNA表达。结果:PRDX Ⅳ mRNA在3周龄、6周龄、8月龄卵巢中表达分别为新生3d小鼠的0.76、0.42、0.12(P<0.05);在性成熟小鼠各动情周期卵巢中表达无差异;在未成熟小鼠促性腺激素预处理卵巢中,PMSG注射48h表达为生理盐水注射组(CON组)的1.63倍(P<0.05),于HCG注射后4、8、12、48h分别为CON组的0.81、0.56、0.78、0.66(P<0.05)。结论:PRDX Ⅳ可能通过调节卵泡发育与卵巢氧化应激压力而调控卵巢衰老过程。
Objective To explore the expression of peroxiredoxin Ⅳ mRNA in the follicle development and ovarian aging of mice. Method C57BL/6j mice in different stages of age and estrous cycle were pretreated with gonadotropin. Then real-time polymerase chain reaction was used to detect the expression of PRDX Ⅳ mRNA. Results The expressions of PRDX Ⅳ mRNA in the ovaries of mice in 3 weeks old, 6 weeks old, and 8 months old were 0.76, 0.42 and 0.12 of those in 3 days old (P 〈 0.05); And were not significantly different among matured mice in different estrous cycle. In immature mice pretreated with gonadotropin, the expression of PRDX at 48 h after PMSG injection was 1.68 of that after normal saline injection (CON group) (P 〈 0.05) ; and those at 4, 8, 12, 48 h after HCG injection were 0.81, 0.56, 0.78, 0.66 of those in CON group (P 〈 0.05). Conclusion PRDX IV may probably regulate ovarian aging through regulating follicle development and ovarian oxidative stress.
