摘要
目的:建立复方两面针含片中苦参碱和没食子酸的HPLC含量测定方法。方法:色谱柱为ZORBAX Eclipse XDB C18(250 mm×4.6 mm,5μm)柱;流动相为乙腈-甲醇-磷酸盐缓冲液(pH 6.8)-三乙胺(16∶16∶68∶0.1)和乙腈-0.1%磷酸0.1%三乙胺水溶液(3∶97),流速1.0 mL.min-1;柱温30℃;检测波长220 nm。结果:苦参碱在0.10~2.00μg范围内线性关系良好(r=0.99960,n=6);平均回收率为99.4%(n=9)。没食子酸在0.011~1.75μg范围内线性关系良好(r=0.99998,n=6);平均回收率为99.1%(n=9)。结论:建立的方法适用于复方两面针含片的质量控制。
Objective: To establish an analytical method for determination of matrine and gallic acid in Compound Liangmianzhen tablets by HPLC.Methods: The analysis was achieved on a ZORBAX Eclipse XDB C18 column(250 mm ×4.6 mm,5 μm) using a mobile phase of acetonitrile-methanol-phosphate buffer(pH 6.8)-triethylamine(16∶ 16∶ 68∶ 0.1) and a mobile phase of acetonitrile-0.1% phosphoric acid and 0.1% triethylamine water solution(3∶ 97) as the mobile phase;the flow rate was 1.0 mL.min-1;the detecting wavelength was 220 nm and the column temperature was 30 ℃.Results: The calibration curve of matrine was linear within the range of 0.10-2.00 μg(r =0.9996,n =6),theaverage recovery was 99.4%(n =9).The calibration curve of gallic acid was linear within the range of 0.011-1.75 μg(r = 0.9999,n = 6),the average recovery was 99.1%(n = 9).Conclusion: The method can be used to control the quality of the Compound Liangmianzhen tablets.
