固态发酵制备矛尾刺鰕虎鱼抗氧化蛋白水解液

Preparation of Antioxidant Protein Hydrolysate Derived from Goby Acanthogobius hasta Muscle by Solid State Fermentation

摘要 研究利用米曲霉固态发酵矛尾刺鰕虎鱼肌肉蛋白制备抗氧化蛋白水解液的发酵条件。在考察蛭石含量、接种量、初始pH、含水率对菌株产蛋白酶及多肽液的1,1-二苯基-2-三硝基苯肼自由基清除率影响的基础上,研究菌株产蛋白酶、平均多肽链长、水解度、1,1-二苯基-2-三硝基苯肼自由基清除率随时间的变化趋势,获得最佳发酵条件。试验结果表明,发酵条件为40%(m/m)直径1.0~1.5mm蛭石、接种量5%(V/m)、初始pH 7.5、含水率55%、30℃、发酵96h后,多肽平均链长为10,1,1-二苯基-2-三硝基苯肼自由基清除率达到68.5%,水解度为32%。 Preparation of antioxidant hydrolysate derived from goby Acanthogobius hasta muscle was optimized under solid state fermentation by yeast Aspergillus oryzae AS 3. 951 to provide the foundation for low cost, and large-scale production of antioxidant hydrolysate. The effects of the vermiculite content, inoculum size, initial pH and moisture content on the production of the protease by A. oryzae and the DPPH free-radical scavenging activity of the hydrolysate of the goby protein, the production of protease, the average peptide length, hydrolysis degree, and DPPH free-radical scavenging activity of the hydrolysate were investigated in the process of solid state fermentation. The fermentation were found to be carried out under the conditions of 40~ (rnJrn)vermiculite, 5 ~ inoculum size, initial pH 7.5, moisture of 55~, 30 *C, and for 96 h. Under those conditions, the average peptide was 10 in length, and hydrolysis degree of 68.5~, and the hydrolysate showed DPPH free-radical scavenging activity of 32~.
