摘要
目的研究苦参碱改构体X体外诱导人鼻咽癌CNE1细胞凋亡的机制。方法用不同剂量改构体X处理CNE1细胞48h,透射电镜观察CNE1细胞超微结构变化;Westernblot检测改构体X对CNE1细胞内Caspase-3,-8,-9蛋白表达的影响。结果苦参碱改构体X作用CNE1 48 h后,细胞出现皱缩,核内异染色质明显增多,核周隙增大,凋亡小体形成等细胞凋亡特征形态,高剂量组较低剂量组细胞凋亡程度更加明显;Western blot检测显示,与阴性对照组相比,除低剂量改构体X对Caspase-8无上调作用外,其他剂量组可上调凋亡相关蛋白Caspase-3,-8,-9的表达(P<0.01),且具有剂量依赖性。结论苦参碱改构体X可通过上调促凋亡蛋白Caspase-3,-8,-9的表达诱导CNE1细胞凋亡。
Aim To study the mechanism of matrine deiwant X inducing apoptosis of human nasopharyngeal carcinoma CNE1 cells in vitro. Methods Transmission electron microscope observation on changes of ultra structure of CNE1 cells was done after CNE1 cells were treated with different dosage of matrine derivant X for 48h, and the protein expression of Caspase-3, -8, -9 was detected by western blotting. Results After incubation of CNE1 cells treated with matrine derivant X for 48h, characteristic morphology of apoptosis appeared, such as cell shrinking, significant increases of in nucleus, increased gap of nucleus periphery, and formation of apoptotic bodies, and the apoptotic characteristics were more visible in high dose group than low dose group. Western blot showed that the expression of Caspase-3, Caspase-$ and Caspase-9 protein increased significantly in a dose-dependent manner in different dosage groups ( P 〈 0.01 ) except Caspase-8 in low dosage group compared with negative control group. Conclusion The matrine derivant X can induce apoptosis of CNE1 cells by increasing the Caspase-3, Caspase-8 and Caspase-9 protein expression.
