解毒活血方对动脉粥样硬化易损斑块兔模型CD40及白细胞介素-1β表达的影响 被引量:10

Effect of Jiedu Huoxue Recipe on CD40 and Interleukin-1 Beta Expression in Rabbit Model of Vulnerable Atherosclerotic Plaque

摘要 【目的】探讨解毒活血方对动脉粥样硬化(AS)易损斑块兔模型CD40、白细胞介素-1β(IL-1β)表达的影响。【方法】采用球囊损伤主动脉内膜联合高脂饲料法复制实验性动脉粥样硬化易损斑块兔模型,造模兔随机分为中药低、高剂量(剂量分别为40、80 g.kg-1.d-1)组,辛伐他汀(剂量为2.6 mg.kg-1.d-1)组,模型对照组,另设正常对照组以生理盐水灌胃,疗程10周。观察主动脉病理改变,检测血清sCD40L(soluble CD40L)、IL-1β及斑块CD40 mRNA、IL-1βmRNA表达。【结果】模型对照组呈易损斑块特点,中药高剂量组呈稳定斑块特征,各干预组均能不同程度降低血清sCD40L、IL-1β表达水平,高剂量解毒活血方能显著降低斑块内CD40 mRNA、IL-1βmRNA表达。【结论】解毒活血方能下调CD40L、IL-1β表达,具有一定稳定易损斑块的作用。 Objective To explore the effect of Jiedu Huoxue Recipe(JHR)on CD40 and interleukin-1 beta(IL-1β)expression in rabbit model of vulnerable atherosclerotic plaque.Methods Rabbit model of atherosclerosis(AS)-induced vulnerable plaque was established by injury of aorta intima with balloon and by high-cholesterol feeding.And then the model rabbits were randomly divided into low-dose JHR(40 g·kg^-1·d^-1)group,high-dose JHR(80 g·kg^-1·d^-1)group,simvastatin(2.6 mg·kg-1·d-1)group,and model control group.Additionally,normal control group was set up and was treated with normal saline.The treatment course lasted 10 weeks.HE-staining aortic pathological changes,serum soluble CD40 ligand(sCD40L)and IL-1β contents as well as plaque CD40 mRNA and IL-1β mRNA expression were measured.Results The model control group was characterized as vulnerable plaque,while high-dose JHR group was characterized as stable plaque.Serum sCD40L and IL-1β contents were decreased in all of the medication groups,and plaque CD40 mRNA and IL-1β mRNA expression levels were significantly reduced in high-dose JHR group.Conclusion JHR can stabilize the vulnerable atherosclerosis plaque by down-regulating the mRNA expression of CD40L and IL-1β.
