盐酸小檗碱对结肠癌HT29细胞鸟氨酸脱羧酶表达的影响 被引量:2

Effect of Berberine Hydrochloride on Ornithine Decarboxylase Expression in Colon Cancer HT29 cells

摘要 【目的】观察盐酸小檗碱对结肠癌HT29细胞鸟氨酸脱羧酶(ODC)表达的影响。【方法】以不同浓度的盐酸小檗碱作用于HT29细胞72 h,采用四甲基偶氮唑盐(MTT)法测定细胞增殖抑制,采用荧光定量PCR法及Western blot方法分别检测ODC mRNA及蛋白的表达水平。【结果】盐酸小檗碱可抑制HT29细胞增殖,剂量依赖性地显著下调ODC蛋白表达(P<0.05或P<0.01),但对ODC mRNA表达无显著影响。【结论】盐酸小檗碱具有抑制HT29细胞的增殖作用,其机制可能与下调ODC蛋白表达相关。 Objective To study the effect of berberine hydrochloride on ornithine decarboxylase(ODC) expression in colon cancer HT29 cells.Methods HT29 cells were treated with various concentration of berberine hydrochloride for 72 hours,and then the proliferation of HT29 cells was detected by methyl thiazolyl tetrazolium(MTT) assay.Real-time fluorescent quantitative PCR was used to quantitatively analyze ODC mRNA expression,and ODC protein expression was detected by Western blotting method.Results Berberine hydrochloride could inhibit the proliferation of HT29 cells,dose-dependently down-regulated ODC protein expression in HT29 cells(P〈0.05 or P〈0.01),but had no influence on ODC mRNA expression.Conclusion Berberine hydrochloride had inhibitive effect on the proliferation of HT29 cells,and the mechanism may be associated with the down-regulation of ODC protein.
