摘要
本文探讨体外培养的各种成纤维细胞在机械力的作用下,细胞骨架、生长方向、细胞增殖、细胞外基质和生长因子发生的变化,以及可能发生的信号传导途径变化。在机械应力作用下,成纤维细胞的形态和骨架发生了规律的改变;细胞的生长方向发生重排,且与力的方向呈垂直趋势;适宜范围内的机械力可以促进细胞的增殖;机械应力的刺激可引起细胞外基质合成增加和多种生长因子的分泌,导致组织适应性改变;而出现这些现象的机制,可能是机械力作用于细胞启动了MAPK、一氧化氮和氧化应激途径。
This paper mainly reviews the studies on the changes of cytoskeleton, cell orientation, pro- liferation, growth {actors, extracellular matrix, as well as the possible signaling pathways of fi- broblasts under the action of mechanical strain in vitro. Studies have found that under the effect of mechanical strain, the morphology and cytoskeleton of fibroblast take regular change. Appro- priate degree of mechanical strain can stimulate fibroblast proliferation and uniform alignment perpendicular to the direction of strain. Mechanical force can also lead to synthesis of extracellu- lar matrix and secretion of a variety of growth factors, which can induce adaptive change in tis- sue. And the possible mechanism of these responses may due to the activation of the MAPK, ni- tric oxide and oxidative stress signaling pathways.
