奥沙利铂热化疗对卵巢癌细胞A2780凋亡及侵袭转移的影响 被引量:4

Effect of oxaliplatin thermochemotherapy on the apoptosis and invasion of human ovarian cancer A2780 cells

摘要 目的探讨热疗、奥沙利铂化疗及奥沙利铂热化疗对卵巢癌细胞A2780凋亡及侵袭转移的影响。方法卵巢癌细胞A2780进行体外培养,分为4组,分别为正常对照组(A组)、热疗组(B组)、奥沙利铂化疗组(C组)与奥沙利铂热化疗组(D组),采用MTT法分别测定4组细胞抑制率,并判断奥沙利铂热化疗联合作用的效果,采用流式细胞仪检测细胞凋亡率,采用反转录PCR检测基质金属蛋白酶-2及血管内皮生长因子mRNA表达情况。结果与B、C组比较,D组细胞抑制率及凋亡率明显增加(P<0.01),基质金属蛋白酶-2及血管内皮生长因子mRNA表达明显降低(P<0.01)。结论奥沙利铂热化疗明显促进卵巢癌细胞株A2780凋亡,抑制其侵袭转移。 Objective To investigate the effect of hyperthermia, oxaliplatin chemotherapy and oxaliplatin thermochemotherapy on the apoptosis and invasion of human ovarian cancer A2780 cells. Methods Human ovarian cancer A2780 cells were cultured in vitro and divided into four groups, including normal control group (group A), hyperthermia group (group B), oxaliplatin chemotherapy group (group C) and oxaliplatin thermochemotherapy group (group D). MTT assay was used to determine the cell inhibition rate, and the effect of oxaliplatin thermochemotherapy was evaluated. Flow cytometry was used to analyze apoptosis rate. Reverse transcription PCR technique was used to observe the expression of matrix metalloprotinease-2 (MMP-2) and vascular endothelial growth factor (VEGF) mRNA. Results The cell inhibition and apoptosis rates increased, and MMP-2 and VEGF mRNA decreased in group D compared with group B and C (P〈0.01). Conclusion Oxaliplatin thermochemotherapy could apparently promote apoptosis of A2780 cells and inhibit invasion of A2780 ceils in vitro.
