白芍中芍药苷提取工艺的对比研究 被引量:36

Comparison of Different Extracting Processes for Paeoniflorin in Paeoniae Radix Alba

摘要 目的:优选白芍中芍药苷的提取工艺条件,为白芍中芍药苷的提取提供依据。方法:将安徽亳州产白芍道地药材分别用水提取、乙醇回流提取、乙醇超声提取、乙醇索氏提取,以芍药苷为指标成分,按《中国药典》2010年版规定项下方法进行测定,优选最佳提取工艺并进行工艺验证;色谱条件为Kromasil 100-5C18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸溶液(14∶86),检测波长230 nm,柱温30℃,进样量10μL,流速1.0 mL.min-1。结果:理论塔板数按芍药苷峰计算均不低于2 000。在此条件下,芍药苷与其他组分分离较好;芍药苷得率依次为50%乙醇超声提取>80%乙醇回流提取>50%乙醇回流提取>80%乙醇索氏提取>水提取。结论:白芍中芍药苷以50%乙醇超声提取为最佳。 Objective: To choose the best extraction for paeoniflorin in Paeoniae Radix Alba from different extracting processes and provide the basis.Method: The medicinal materials from original growing areas were extracted by water,enthanol reflux,enthanol ultrasonic extracting and enthanol Soxhlet extracting respectively.The best extracting process was chosen from different extracting processes based on determining the content of index components of paeoniflorin by HPLC according to Chinese Pharmacopoeia 2010 and verified three times.A Kromasil 100-5C18 analytical column(4.6 mm × 250 mm,5 μm) was used.The mobile phase was consisted of acetonitrile and 0.1% H3PO4(14∶ 86);the detection wavelength was set at 230 nm.The flow rate was 1.0 mL.min-1 and the column temperature was kept at 30℃.The sample size was 10 μL.Result: The determining condition of paeoniflorin was good according to Chinese Pharmacopoeia 2010.The content of paeoniflorin in Paeoniae Radix Alba was 3.02% for 50% enthanol ultrasonic extracting,2.90% for 80% enthanol reflux extracting,2.64% for 50% enthanol reflux extracting,1.83% for 80% enthanol Soxhlet extracting and 1.56% for water extracting respectively.Conclusion: The best extraction for paeoniflorin in Paeoniae Radix Alba from different extracting processes is 50% enthanol ultrasonic extracting.
