摘要
通过液体培养,跟踪检测了蝉棒束孢LB菌株在培养过程中孢子萌发、菌丝干重、pH、还原糖含量、氨基氮含量等培养参数。结果表明,随培养时间延长,培养液中还原糖含量、氨基氮含量、酸碱度和菌丝生物量均发生明显的变化。接种培养48 h内,菌丝产量增长较慢;培养48~144 h,菌丝生物量增长幅度最大;培养168 h,菌丝生物量达22.45 g/L。培养液的pH、糖含量、氮含量随培养时间的延长逐渐降低;培养72 h后营养液的pH降为4.56,因此培养72 h后,可适当增加pH,以利蝉棒束孢菌丝生长。
Growing indexes of Isaria cicadae strain LB such as pH value, mycelia biomass, contents of reducing sugar and amino nitrogen were tested systemically. The results showed that contents of reducing sugar, amino nitrogen, pH values and mycelia biomass in nutritive medium changed obviously in the process of inoculation. The mycelial biomass increased rapidly in early 48-144 h and reached 22.45 mg/mL after 168h. The pH value, contents of reducing sugar and amino nitrogen de- creased gradually in the course of submerged culture of I. cicadae, and initial pH value of the medium dropped from 5.63 to 4.42 in 72 h after being inoculated. For mycelia growth, pH value could be regulated reasonably in 72 h after being inoculated.
