盐酸埃克替尼对人舌鳞癌Tca-8113细胞凋亡及Bcl-2和Bax蛋白表达的影响 被引量:6

摘要 目的研究表皮生长因子受体酪氨酸激酶抑制剂盐酸埃克替尼对人舌鳞状细胞癌Tca8113细胞凋亡及Bcl-2、Bax蛋白表达的影响,探讨其诱导肿瘤细胞凋亡的可能机制。方法分别应用0、10、20、40μmol.L-1盐酸埃克替尼处理体外培养的人舌鳞状细胞癌Tca8113细胞,分别作用24、48、72 h后,采用流式细胞仪测定细胞凋亡率,采用间接免疫荧光联合流式细胞技术及免疫细胞化学方法检测盐酸埃克替尼对人舌鳞癌Tca8113细胞Bcl-2、Bax蛋白表达的影响。结果盐酸埃克替尼作用于人舌鳞癌Tca8113细胞后Bcl-2蛋白表达降低,Bax蛋白表达增加;随盐酸埃克替尼浓度、作用时间的增加,人舌鳞癌Tca8113细胞凋亡率增加;人舌鳞癌Tca8113细胞凋亡率与盐酸埃克替尼浓度、作用时间成正相关。结论盐酸埃克替尼可降低Bcl-2蛋白表达,增加Bax蛋白表达,诱导人舌鳞癌Tca8113细胞凋亡。 Objective To study the effect of icotinib hydrochloride on the apoptosis of human carcinoma of tongue Tca8113 cell line and the expressions of Bcl-2 and Bax protein in order to explore the mechanisms.Methods Tca8113 cells were incubated with icotinib hydrochloride at a dose of 0,10,20,40 μmol·L-1 for 24,48,72 h respectively.Apoptosis were detected by flow cytometer(FCM).Immunocytochemical method,indirectly immunofluorescence and FCM were used to detect the expression of the Bcl-2 and Bax protein after Tca8113 cells were treated with icotinib hydrochloride.Results After the treatment of icotinib hydrochloride,the Bcl-2 protein expression decreased and the Bax protein expression increased of human carcinoma of tongue Tca8113 cells.The percentage of apoptotic cells was gradually increased with the increasing of concentration and treating time of icotinib hydrochloride.The percentage of apoptotic cells were positive correlated to the concentration and treated time of icotinib hydrochloride.Conclusion Icotinib hydrochloride can decrease the expression of the Bcl-2 protein and increase the expression of the Bax protein,and it can induce apoptosis of human carcinoma of tongue Tca8113 cell.
