摘要
采用栓线法和四动脉结扎法造成大鼠脑缺血再灌注模型 ,观察了海洋硫酸多糖类药物“989”对缺血脑组织的保护作用。结果发现 ,“989”在 2~ 10 mg·kg-1范围内静脉注射 ,能明显缩小脑栓塞大鼠 2 4 h的脑梗塞范围 ,改善行为障碍 ,对局灶性脑缺血和全脑缺血均有明显治疗作用。进一步研究发现 ,其作用机制可能与抑制脑组织 TNF- α表达有关。
The protective effects of 989,a kind of marine polysaccharides extracted from marine living organisms by enzymatic fractionation and specific modification,against damage induced by transient focal and global ischemic Wistar rats were studied in this paper.Wistar rats at body weight ranging from 300~400g were randomly divided into desired groups.The transiet focal and global cerebral ischemia models were successfully established by modified methods to evaluate the effects of our detected chemical agent.The results indicated that 989 treated intravenously at doses ranging from 2~10 mg·kg 1 reduced cerebral infarcted area, characterized by promoting the behavioral deficit in focal ischemia reperfusion model.Futhermore,989 at doses mentioned above enhanced neuronal survival in global cerebral ischemic rats.The antiischemic activity exerted by 989 was supposed to contribute to its neuronal survival-promoting activity and also to its inhibitory effect against the production of tumor necrosis factor(TNF α).
